Selective ligand activity at Nur/retinoid X receptor complexes revealed by dimer‐specific bioluminescence resonance energy transfer‐based sensors

Abstract: Retinoid X receptors (RXRs) play a role as master regulators because of their capacity to form heterodimers with other nuclear receptors (NRs). Accordingly, retinoid signaling is involved in multiple biologic processes, including development, cell differentiation, metabolism, and cell death. However, the role and function of RXRs in different heterodimer complexes remain unidentified, mainly because most RXR drugs (called rexinoids) are not selective of specific heterodimer complexes. The lack of selectivity s… Show more

“…It is thus possible that its association with reproductive lifespan was driven by the effect on AM in EAs and the effect on ANM in AAs. This SNP is intronic in RXRG , which encodes retinoid X receptor gamma, a nuclear receptor that dimerizes with the receptors for retinoic acid, thyroid hormone and vitamin D and increases their DNA binding and transcriptional activity[ 47 ]. However, the biological impact of the SNP on gene expression and the mechanism of RXRG in regulating reproductive aging, remain unclear.…”

“…Clobetasol-dependent MBP gene expression and the morphological changes accompanying Oli-neuM differentiation upon Clobetasol treatment require Smo activation and RxRγ gene transcription as cyclopamine or itraconazole treatment reduces MBP expression in Clobetasol-treated Oli-neuM and RxRγ inhibition in Clobetasol-treated Oli-neuM results in MBP downregulateion [ 15 ]. RxRγ can form homodimers or heterodimers with other nuclear receptors (RAR, RxR, VxD or PPARs) depending on the ligand stimulation [ 65 ]. RxRγ gene expression is upregulated at remyelination lesions in MS patients and its downregulation in cultured OPCs results in depletion of differentiated OLs in purified OPC cultures, with OLs stalled at the premyelination stage.…”

“…All the PEC measurements were performed with a 250 W Xe lamp (Beijing Trusttech Co. Ltd.) as the visible-light source (passing through a 400 nm UV-cut filter) at the intensity of 100 mW cm −2 . The electrochemical impedance spectroscopy (EIS) was performed with a ZENNIUM electrochemical workstation (Zahner Instruments, Germany) in 0.10 M KCl solution containing 5.0 mM Fe(CN) 6 3−/4− .…”

“…Recently, various types of DNA sensors or aptasensors have been fabricated on the basis of RET involving fluorescence RET [4], chemiluminescence RET [5], bioluminescence RET [6] and electrochemiluminescence RET [7][8][9]. As we all know, the key to promoting the detection sensitivity of the RET-based sensor is to improve the efficiency of RET between energy donor and acceptors, which is highly dependent on the spectral overlap between the donor's emission and the acceptor's absorption [10,11].…”
