A simple and highly sensitive electrochemical method, based on a gold nanoparticle (GNP)-modified indium tin oxide (ITO) electrode (ITO/GNP), was developed for the detection of melamine (MLM). The modified electrode surface was characterized by scanning electron microscope (SEM) and cyclic voltammetry (CV). MLM was detected based on the adsorption of MLM onto ITO/GNP electrode causing suppression in the cathodic peak current of ferricyanide. A wide linear concentration range from 5 to 500 nM with a lower detection limit of 1.0 nM (i.e. 0.13 ppb) for MLM was obtained. The proposed method was successfully applied to detect MLM in milk samples. It has been illegally added to dairy products by unethical manufacturers to artificially increase the apparent protein content. 2 High concentration of MLM can induce acute kidney failure, serious renal problems and even death. 3 Therefore, a highly sensitive analytical method is necessary for the determination of MLM in dairy products.Various methods, such as high performance liquid chromatography, capillary electrophoresis, chemiluminescence, and electrochemical techniques have been reported for the determination of MLM. 4 Electrochemical methods are more advantageous than the others mostly due to fast response, low cost, simple operation, and high sensitivity. 5,6 However, the problem with this method is the electrochemical inactivity of the MLM. Therefore, MLM can be detected indirectly by applying electrochemical method. Few reports are available regarding the indirect electrochemical detection of MLM. Cao et al. applied the interactions between oligonucleotides and MLM lead to the increase in the peak currents of ferricyanide at an oligonucleotide-modified gold electrode for electrochemical sensing of MLM. 4 The same group further reported an electrochemical sensor for MLM with 3,4-dihydroxyphenylacetic acid as the recognition element. 7 In this study, we have used ferricyanide as the recognition element and gold nanoparticle (GNP) modified indium tin oxide (ITO) electrode as base materials. MLM was attached with the ITO/GNP electrode through strong interaction between the -NH 2 group of MLM and GNP. After the attachment of MLM, the cathodic peak current of ferricyanide was found to suppress, which is directly related with the concentration of MLM and can be used to build up an electrochemical detection scheme for the determination of MLM. ExperimentalAll reagents were obtained as analytical grade and used without further purification. Double distilled water was used throughout the experiments. A novel and convenient electrochemical method has been developed for sensitive determination of melamine (MEL) using ascorbic acid (AA) as the recognition element. The working electrode employed in this method was modified with the nanocomposite of hydroxyapatite/carbon nanotubes to enhance the current signal of recognition element. The interaction between MEL and AA was investigated by fourier transform infrared spectroscopy and cyclic voltammetry, and the experimental results indicated that hydrogen bonding was formed between MEL and AA. Because of the existing hydrogen bonding and electrostatic interaction, the anodic peak current of AA was decreased obviously while the non-electroactive MEL added in. It illustrated that the MEL acted as an inhibitor to the oxidation of AA and the decreasing signals can be used to detect MEL. Under the optimal conditions, the decrease in anodic peak current of AA was proportional to the MEL concentrations ranging from 10 to 350 nM, with a detection limit of 1.5 nM. Finally this newly-proposed method was successfully employed to detect MEL in infant formula and milk, and good recovery was achieved.

Sensitive colorimetric detection of melamine with 1,4-dithiothreitol modified gold nanoparticles

A colorimetric detection method of melamine based on 1, 4-dithiothreitol (DTT) functionalized gold nanoparticles (DTT-AuNPs) is reported. Hydrogen-bonding interaction between DTT and melamine resulted in the aggregation of AuNPs, consequent color changes of AuNPs from wine red to blue and red-shift of the surface plasmon resonance (SPR) peak of DTT-AuNPs. The concentrations of melamine could be determined with naked eye or a UV-vis spectrometer. FT-IR and TEM were used to investigate the modification of DTT-AuNPs. Results showed that the absorption ratio (A 680 /A 520 ) was linear with the logarithm of melamine concentration in the range of 8.0×10 -8 M to 6.0×10 -7 M and 6.0×10 -7 M to 1.5×10 -6 M with linear coefficients of 0.993 and 0.999, respectively. The detection limit (S/N = 3) of the proposed method was 2.4×10 -8 M, which was much lower than the safety limit. The coexisting substances did not affect the determination of melamine. Furthermore, the proposed method was applied for detecting trace melamine in real milk samples, with recoveries of 96%-103%.
