“…Due to its antimitotic activity, it is easy to understand that topical 5-FU is a useful therapy for the treatment of many dermatological disorders characterized by a high mitotic rate (e.g., nonmelanoma skin cancers, actinic keratosis, benign tumours, nail psoriasis, and porokeratosis) [1012]. In the same way, it is expected that 5-FU applied to a wound will strongly inhibit wound epithelialization and, consequently, will delay the wound healing [13], so it is surprising that 5-Fu would be implicated in the vitiligo repigmentation process which obviously needs melanocyte proliferation.…”

“…The Mohs chemosurgical technique has also been reported to have a high cure rate 15,16 . More recently, therapy with topical 5FU cream has been advocated for the treatment of EQ, with favorable results 3–8 . This has been preferred because of its ability to selectively destroy tumor cells without significant damage to adjacent normal tissues.…”

“…Although EQ has been expanded to involve CIS of all mucosal structures of men and women, Graham recommends the strict limitation of this concept to the male genitalia 3 . The high recurrence rate as well as the potentially mutilating effects of conventional treatment had led investigators to use topical 5‐fluorouracil (5FU) in an attempt to conservatively ablate this potentially invasive form of CIS 4–6 . Subsequent reports have confirmed the effectiveness of this modality for the treatment of EQ 7,8 …”

“…Topical FU can be extemporaneously prepared by the pharmacist on prescription, and such a preparation has a number of advantages which would recommend its routine use. In the approximately five years from the time topical FU was first introduced as an effective therapy for multiple actinic keratoses by Dillaha et al (5) until it became commercially procurable, only extemporaneous preparations were available. Those of us who early embraced this form of therapy gained familiarity with it and knowledge of expected reactions; accordingly we continued to use it after commercial preparations became available.…”
