Selection of Nucleic Acid Aptamers Specific for Mycobacterium tuberculosis

Abstract: Tuberculosis (TB) remains to be a major global health problem, with about 9 million new cases and 1.4 million deaths in 2011. For the control of tuberculosis as well as other infectious diseases, WHO recommended "ASSURED" (Affordable, Sensitive, Specific, User-friendly, Rapid and robust, Equipment-free, and Deliverable to the end user) diagnostic tools that can easily be maintained and used in developing countries. Aptamers are promising tools for developing point-of-care diagnostic assays for TB. In this stud… Show more

“…The SELEX (systematic evolution of ligands by exponential enrichment) technique was generated by using reiterative in vitro selection for high-a nity and -speci city oligonucleotide ligands (aptamers) against almost any kind of molecules that are of biological or therapeutic interest [35]. Aptamers provide a powerful tool not only used for identi cation of novel diagnostic markers but also for interference with the duplication of the pathogenic agents, and thus have been widely used as biorecognition probes against human pathogens and/or their toxins [36][37][38].…”

“…Another aptamer, MTB36 also shows preferential binding with M.tb over M . bovis BCG suggesting its ability to discriminate between close species ( Mozioglu et al., 2016 ). PPK (polyphosphate kinase gene) regulates inorganic polyphosphate (polyP) function and aids in virulence.…”

“…Multiple aptamers have been developed that target strains of this bacterium or virulence factors for its havoc in humans. Mozioglu et al isolated the H37Ra strain as the positive target in their SELEX study [ 33 ]. Both ultrafiltration and centrifugation methods were used to partition the bound and unbound library molecules.…”
