SummaryThe expanding aquaculture industry plays an important role in feeding the growing human population and with the expansion, sustainable bacterial disease control, such as probiotics, becomes increasingly important. Tropodithietic acid (TDA)‐producing Phaeobacter spp. can protect live feed, for example rotifers and Artemia as well as larvae of turbot and cod against pathogenic vibrios. Here, we show that the emerging live feed, copepods, is unaffected by colonization of the fish pathogen Vibrio anguillarum, making them potential infection vectors. However, TDA‐producing Phaeobacter inhibens was able to significantly inhibit V. anguillarum in non‐axenic cultures of copepod Acartia tonsa and the copepod feed Rhodomonas salina. Vibrio grew to 106 CFU ml−1 and 107 CFU ml−1 in copepod and R. salina cultures, respectively. However, vibrio counts remained at the inoculum level (104 CFU ml−1) when P. inhibens was also added. We further developed a semi‐strain‐specific qPCR for V. anguillarum to detect and quantify the pathogen in non‐axenic systems. In conclusion, P. inhibens efficiently inhibits the fish larval pathogen V. anguillarum in the emerging live feed, copepods, supporting its use as a probiotic in aquaculture. Furthermore, qPCR provides an effective method for detecting vibrio pathogens in complex non‐axenic live feed systems.

In the present study, two strains of Malaysian Paenibacillus spp. isolated from intestinal tract of cultured hybrid red tilapia were tested for their antagonistic activity against Vibrio alginolyticus, a destructive bacterial pathogen to marine fishes as probiotics in aquaculture. Homology searches of 16S rRNA and Internal Transcribes Spacer (ITS) genes sequencing with nucleotide Basic Local Alignment Sequence Tool (nBLAST) revealed that both strains were identical to Paenibacillus spp. and Paenibacillus pabuli, respectively. In vitro and in vivo experiments were conducted to study the efficacy of these Paenibacillus strains as probiotics. Co-culture assay demonstrated that the growth of V. alginolyticus was inhibited when grown with these probionts. A complete inhibition of the pathogen by P. pabuli strain D12 occurred at 48 h of incubation period. To qualify strains as probiotics, Artemia challenged with V. alginolyticus was performed, with or without these potential probiotics. The highest survival rate (72%) of Artemia was observed in P. pabuli strain D12 treatment after challenged with V. alginolyticus, followed by P. pabuli strain D14 at 68% survival rate. The lowest survival of Artemia (23% survival rate) was recorded when challenged with only V. alginolyticus. This study reported the ability of Paenibacillus D12 and D14 as potential probiotics to control V. alginolyticus in Artemia culture system.

The identification and use of probiotic bacterial stains is a practical approach to protect clams grown in aquaculture farms from disease. The inhibition of the pathogenic bacterium Vibrio alginolyticus was used as a trait to select a candidate probiotic bacterial strain in this study. An ideal bacterial strain, SW-1, was isolated from seawater from a clam farm. The selected isolate SW-1 was identified based on its physiological, morphological, and biochemical characteristics and its 16S rDNA sequence. The experiments showed that strain SW-1 had a high similarity to Pseudoalteromonas piscicida and could inhibit the growth of V. alginolyticus (V.-MP-1). SW-1 also improved the survival of clams following challenge with the pathogenic V.-MP-1. The mortality of clams was 100% after infection with 10 8 CFU/mL of V. alginolyticus, whereas mortality was only 11% when clams were infected with 10 8 CFU/mL of V.-MP1 while simultaneously exposed to the same concentration of Pseudoalteromonas SW-1, indicating that Pseudoalteromonas SW-1 could be used as a probiotic to protect farmed clams, and thus reduce the effects of antibiotics on aquatic environment.
