dBiochar (BC) amendments might decrease the bioavailability of metals in soils that are contaminated with heavy metals. In general, soil microbial communities are sensitive to changes in soil property changes. Microbial communities were tested in a Cd-and Pb-polluted paddy field in southern China. BC was applied as a basal soil amendment before rice transplantation in 2009. The BC was applied at rates of 0, 10, 20, and 40 tons per hectare. Soil heavy metal fractions with sequential extraction procedure, soil microorganisms, and enzymes were monitored in 2011. The soil pH and soil organic carbon (SOC) were significantly increased by 2% to 5% and 16% to 51% under BC amendment, respectively. Compared to the non-BC treatment, the cadmium (Cd) and lead (Pb) acid-soluble fraction concentrations were significantly decreased by 15.3% to 26.7% and 18.2% to 30.9%. The Cd and Pb reducible fraction were decreased by 13.5% to 25.6% and 21.9% to 23.53%.The Cd and Pb oxidizable fraction by 15.4% to 69.2% and 22.7% to 29.3% with BC application, respectively. The populations of actinomycetes and fungi were increased by 19.0% to 38.5% and 3.7 to 9.3 times, respectively. Meanwhile, BC significantly increased the cellulose, urine enzyme, neutral phosphatase, and sucrase activities by 117.4% to 178.3%, 31.1% to 37.6%, 29.7% to 193.8%, and 36.5% to 328.6%, respectively. BC amendment offers a basic option to reduce Cd and Pb bioavailability and change the fractions. The BC also increases microorganism quantity and soil enzyme activity.

The effects of cadmium (CdCl2·7H2O) on cellulase, urease, amylase, invertase and phosphatase were assessed for a period of 45 days in the presence and absence of earthworms [Lampito mauritii (Kinberg)] in alfisol soil. The activities of all enzymes significantly increased with longer incubation times (45 days) under laboratory conditions in both control and Cd-amended soils (both with and without earthworm incubation). However, the activities of all enzymes decreased with increasing Cd concentrations under laboratory conditions, both in the presence and absence of earthworms. In the presence of earthworms, cellulase, urease, invertase and amylase activities increased. However, phosphatase activity was lower in most of the Cd-amended soils in the presence of earthworms compared to its activity levels in soils lacking earthworms. These results show that earthworms modulated the stress imposed by Cd by providing suitable substrates, which in turn acted as stimulants for extracellular enzyme secretion by microbes, and by removing Cd through its accumulation in the tissues of the earthworms.

The straw can be degraded efficiently into humus by powerful enzymes from microorganisms, resulting in the accelerated circulation of N,P,K and other effective elements in ecological system. We isolated a strain through screening the straw degradation strains from natural humic straw in the low temperature area in northeast of china, which can produce cellulase efficiently. The strain was identified as Pseudomonas mendocina by using morphological, physiological, biochemical test, and molecular biological test, with the functional clarification on producing cellulase for Pseudomonas mendocina for the first time. The enzyme force constant Km and the maximum reaction rate (Vmax) of the strain were 0.3261 g/L and 0.1525 mg/(min.L) through the enzyme activity detection, and the molecular weight of the enzyme produced by the strain were 42.4 k D and 20.4 k D based on SDS-PAGE. The effects of various ecological factors such as temperature, pH and nematodes on the enzyme produced by the strain in the micro ecosystem in plant roots were evaluated. The result showed that the optimum temperature was 28 C, and the best pH was 7.4»7.8, the impact heavy metal was Pb 2C and the enzyme activity and biomass of Pseudomonas mendocina increased the movement and predation of nematodes.
