Select Flavonoids and Whole Juice From Purple Grapes Inhibit Platelet Function and Enhance Nitric Oxide Release

Abstract: Background-Moderate red wine consumption is inversely associated with coronary ischemia, and both red wine and purple grape juice (PGJ) contain flavonoids with antioxidant and antiplatelet properties believed to be protective against cardiovascular events. Acute cardiac events are also associated with decreased platelet-derived nitric oxide (NO) release. In this study, the effects of PGJ and PGJ-derived flavonoids on platelet function and platelet NO production were determined. Methods and Results-Incubation o… Show more

“…In addition to the inhibition of the extent of aggregation, flavonoids significantly reduced the initial rate of platelet aggregation. These results are consistent with previous reports in which (Freedman et al, 2001;Son et al, 2004;El Haouari et al, 2006;Ghayur et al, 2011;El Haouari and Rosado, 2011;Ro et al, 2015;Liang et al, 2015;Lu et al, 2016). There are marked differences in the antiplatelet effect between genins and heterosidic flavonoids.…”

“…In these in vitro systems the biological activity of red wine and dark chocolate can be accounted for by procyanidins/proanthocyanidins as they induce endothelium-dependent vasodilatation, suppress endothelin-1 synthesis, and inhibit platelet aggregation at low μmol/l or nmol/l concentrations 18 22 25 27…”

“…Although the mechanisms underlying fructose-induced hypertension are not completely established, it has been proposed that increase in blood pressure in fructose-fed rats is secondary to the development of insulin resistance and hyperinsulinemia. 1,9) The flavonoids are a large group of plant-derived compounds that are known to exhibit a lot of biological effects including reducing plasma levels of low density lipoprotein, 11) scavenging reactive oxygen species, 12) inhibiting platelet aggregation, 13,14) and inhibiting carcinogenesis. 15) In addition, a number of flavonoids have been reported to dilate vascular smooth muscle and then reduce blood pressure in various animal models of hypertension.…”
