Segmented Flow Strategies for Integrating Liquid Chromatography–Mass Spectrometry with Nuclear Magnetic Resonance for Lipidomics

Abstract: Building an accurate lipid inventory relies on coordinated information from orthogonal analytical capabilities. Integrating the familiar workflow of liquid chromatography (LC), high-resolution mass spectrometry (HRMS), and tandem mass spectrometry (MS/MS) with proton nuclear magnetic resonance spectroscopy (1H NMR) would be ideal for building that inventory. For absolute lipid structural elucidation, LC–HRMS/MS can provide lower-level structural information with superior sensitivity, while 1H NMR can provide i… Show more

“…For remaining unknown features for which current in silico and data searching strategies yield no results, collecting nuclear magnetic resonance (NMR) data may be possible with our 2D approach, especially if the second-dimension separation were scaled up to match the semipreparative first dimension. Low mM concentrations are typically required for 1 H NMR identification of unknown compounds, but improvement in instrument sensitivity has yielded promising results for lipid identification using analytical-scale LC-MS . Preconcentration of semipreparative fractions may allow even moderate abundance metabolites to produce sufficient NMR signal for structural elucidation.…”

“…The integration of liquid chromatography (LC) with high-resolution mass spectrometry (HRMS), and tandem mass spectrometry (MS/MS) is a powerful tool to analyze or identify a wide range of compounds in various complex mixtures [7,54]. The chromatographic method needs to be optimized for the satisfactory separation of compounds, resulting in the best-quality LC-MS data.…”
