Seeding of mesenchymal stem cells into inner part of interconnected porous biodegradable scaffold by a new method with a filter paper

Abstract: An appropriate physical support provided by scaffolds creates a supportive environment that directs proliferation and differentiation of stem cells. However, it is difficult to homogenously inoculate stem cells into the inner part of scaffolds at high cell densities. In this study, mesenchymal stem cells were seeded into a hydroxyapatite/poly (D, L-lactic-co-glycolic acid) (HAP/PLGA) scaffold that had enough mechanical strength and porous 3-D structure. With an aid of a filter paper placed under the bottom of … Show more

“…The PLGA-based solid scaffolds (LA:GA = 75:25, Mw. 25 kDa; PLGA scaffold, GC Dental Product Co. Ltd.), with a porosity of 80%, were prepared as previously described (Akita et al, 2014 ; Yamanaka et al, 2015 ). Briefly, PLGA scaffolds were resized to ~2 × 3 × 1 mm 3 and soaked in 70% ethanol for 30 min to improve wetting, and then a vacuum pump was used to remove air bubbles.…”

“…We had previously demonstrated the effectiveness of a poly(d,l-lactic-co-glycolic acid) (PLGA) scaffold in the correction of fenestration bone defects created in mandibular alveolar bone in vivo (12,13). PLGA, a biodegradable polymer with excellent biocompatibility, has a higher structural integrity compared to soft scaffolds made from hydrogels and collagen sponge (7,(18)(19)(20)(21)(22)(23). Transplantation of a PLGA scaffold into the fenestration cavity allowed the space to become filled with connective tissue, while no inflammation was observed during the 5-week experimental period.…”

“…Cell proliferation results indicate that the bovine teeth scaffold is suitable for the cell environment and supports cell growth and cell proliferation, and differentiation [18,31]. Yamanaka et al (2015) research results that MSCs were fed into HAP / PLGA blocks for 12 hours, and high seeding efficiency results (up to 90%) could be achieved for a short time of 12 hours [32]. SEM analysis shows that a formation has appeared on the surface of the bovine teeth scaffold with a diameter of 4,864 μm, which indicates exposed dentinal tubules, thus providing a channel for releasing proteins, growth factors, and cell adhesions [33].…”
