Orchids are valued as ornamental plants, bioindicators, and medicinal plants, which implies that some species may be over-collected. Some inhabit very fragile environments and are under threat by the misuse of habitats and anthropogenic impacts. The search for beautiful plants and flowers has increased the number of facilities for micropropagation either by seeding or by cloning plants using in vitro techniques. However, not all species have appropriate media for growth and development that would help in conservation efforts. Cyrtopodium aliciae is an endemic species of rupestrian grassland in Brazil t. It has appeal as an ornamental plant or for use in hybridisation programs dueo its small size and white brownish-purple dotted flowers. This study compared three different media, namely ½ concentration Murashige and Skoog (MS), Vacin and Wendt, and Knudson C, during plant growth and their effect on the acclimatization of Cyrtopodium aliciae. The number and length of shoots and roots, increase in mass, and survival in vitro and ex vitro were analyzed. The experiment was conducted as completely random with a factorial arrangement of treatments (3 × 3) with 10 repetitions per treatment containing 10 plants for the in vitro experiment and 3 repetitions of 10 plants for the ex vitro experiment. Cyrtopodium aliciae performed better in the ½ concentration MS medium with a higher increase in mass, plant development, and survival under both in vitro and ex vitro conditions.

Waitea circinata (Warcup & Talbot) is an orchid antagonist mycorrhizal fungus with biocontrol potential against rice pathogens. This study aimed to optimize the extraction method, obtain a new extract and evaluate its efficiency against rice pathogens in vitro and in vivo, as well as to compare it with other extraction methods and W. circinata. The extracts were obtained and screened for in vitro growth inhibition against the pathogens Cochliobolus miyabeanus, Monographella albescens and Sarocladium oryzae, using the following extracts: mycelial, crude, lyophilized and mycelial mass. An additional in vitro assay was performed with the principal rice pathogen (Magnaporthe oryzae), in order to evaluate the conidial germination and appressorium formation. Based on this evaluation, the lyophilized and mycelial mass extracts were tested in vivo against rice blast (M. oryzae) and compared to the W. circinata mycelial suspension, in different application forms (simultaneous and previous). The mycelial mass extract inhibited all the pathogens, and the crude and lyophilized extracts inhibited C. miyabeanus and M. albescens, respectively. The mycelial mass extract inhibited the M. oryzae conidial germination and appressorium formation by 80 %, and the simultaneous and previous applications suppressed the rice blast by 94 %. These results indicate that the new extract can be used to control rice pathogens.

Wild bananas are believed to have genes for resistance to biotic and abiotic stress in nature, making them potential genetic resources for creating superior varieties. Wild banana seeds, such as Musa acuminata var. tomentosa are generally difficult to germinate in vivo, so that in vitro embryo culture technique is needed. This study aimed to increase embryo germination and regeneration of wild banana M. acuminata var. tomentosa by soaking the seeds as hydropriming. The treatment comprised of soaking the seeds in sterile distilled water for four periods of time: 0 (control), 1, 4, and 7 days. A total of 45 embryos for each treatment were planted on petri dishes containing MS + 0.5 mg/L BA + 1 mg/L biotin + 1 mg/L proline. The results showed that hydropriming increased the rate of embryo germination and regeneration. Seeds soaked for 1, 4, and 7 days successfully resulted in embryo germination percentages of 87%, 62%, and 62%, respectively, while the control unsoaked seeds germinated with a lower percentage of 42%. One-day soaking treatment was the most optimal treatment to increase the rate of germination and regeneration as well as obtained the best vigor as demonstrated by the highest average height of plantlets, number of leaves, and roots than other treatments. Thus, 1-day seed hydropriming is the best treatment for embryo rescue and regeneration of wild banana M. acuminata var. tomentosa
