Secretion of Thioredoxin Peroxidase Protein of Cat Liver Fluke Opisthorchis felineus during Modeling of Experimental Opisthorchiasis

Abstract: Mechanisms of thioredoxin peroxidase secretion by Opisthorchis felineus were studied in vivo and in vitro. Specific antibodies were obtained and used for western blotting and immunohistochemical detection in Syrian hamster model of opisthorchiasis. Secreted thioredoxin peroxidase protein was accumulated in the worm incubation medium under conditions of oxidative stress and in bile duct cells of hamsters with chronic opisthorchiasis.

“…thioredoxin and TPx, can prevent apoptosis (Sripa et al ., 2012 a ). Opisthorchis viverrini - and O. felineus -derived substances are internalized by the bile duct epithelium (Chaiyadet et al ., 2015; Petrenko et al ., 2017; Pakharukova et al ., 2019 c ). Cholangiocytes in culture can capture exosomes of O. viverrini , which is followed by elevated cell proliferation, IL-6 expression and pronounced alterations of the cell phenotype (Chaiyadet et al ., 2015).…”

“…5 ). Proposed mechanisms behind the carcinogenicity of liver fluke infection include biliary-epithelium damage by parasites, long-lasting immune-system–mediated pathogenesis, and effects of parasite-derived substances that are internalized by the bile duct epithelium ( Pakharukova et al, 2019b ; Petrenko et al, 2017 ; Smout et al, 2015 ).…”

“…Some O . felineus products are highly immunogenic [11], they can interact with the epithelium and accumulate inside the cells [10]. It should be also noted that liver fluke-induced CCA is a multifactorial etiology and other factors, such as Helicobacter infection within the biliary tract or presence of carcinogens and their precursors in food might enhance CCA development [26].…”
