Secreted Frizzled Related Proteins in Cardiovascular and Metabolic Diseases

Abstract: Abnormal gene expression and secreted protein levels are accompanied by extensive pathological changes. Secreted frizzled related protein (SFRP) family members are antagonistic inhibitors of the Wnt signaling pathway, and they were recently found to be involved in the pathogenesis of a variety of metabolic diseases, which has led to extensive interest in SFRPs. Previous reports highlighted the importance of SFRPs in lipid metabolism, obesity, type 2 diabetes mellitus and cardiovascular diseases. In this review… Show more

“…[43][44][45] SFRP1, a member of the secreted frizzled-related glycoprotein family that act as extracellular signaling ligands and modulators of Wnt/β-catenin signal transduction pathways, is ubiquitously expressed and has been implicated in cardiac remodeling and fibrosis. 46,47 A nested case-control study from a cohort of middle-aged women demonstrated increased blood concentrations of SFRP1 to be a predictor of cardiovascular events. 48 The principal strengths of our study include the prospective cohort design of the CHS, with adjudicated IS events and subtypes.…”

“…Similarly, the expression of Sfrp2 , another Wnt signaling pathway component was greatly enhanced in E16.5 cKO hippocampi and VZ/SVZ of neocortices ( Figure 5M-5O ). Although members of the SFRP family were first reported as Wnt inhibitors, Sfrp2 regulates anteroposterior axis elongation, optic nerve development, and cardiovascular and metabolic processes by the promoting or inhibiting Wnt signaling pathway(Guan et al, 2021; Marcos et al, 2015; Satoh, Gotoh, Tsunematsu, Aizawa, & Shimono, 2006; von Marschall & Fisher, 2010),…”

“…Previous studies have shown that expression of SFRP4 in human ventricular myocardium correlates with apoptosis-related gene expression [ 37 ]. SFRP4 has been detected during cardiovascular maturation and in the adult heart.…”
