Secondary Sulfonamides as Effective Lactoperoxidase Inhibitors

Abstract: Secondary sulfonamides (4a–8h) incorporating acetoxybenzamide, triacetoxybenzamide, hydroxybenzamide, and trihydroxybenzamide and possessing thiazole, pyrimidine, pyridine, isoxazole and thiadiazole groups were synthesized. Lactoperoxidase (LPO, E.C.1.11.1.7), as a natural antibacterial agent, is a peroxidase enzyme secreted from salivary, mammary, and other mucosal glands. In the present study, the in vitro inhibitory effects of some secondary sulfonamide derivatives (4a–8h) were examined against LPO. The obt… Show more

“…The next paper in the special issue [ 45 ] investigates another enzyme, lactoperoxidase, for its interaction with sulfonamides incorporating (poly)acetoxybenzamide and/or (poly)hydroxybenzamide scaffolds. These secondary sulfonamides were effective-medium potency lactoperoxidase inhibitors, with inhibition constants varying between the nano- to the micromolar range [ 45 ].…”

“…For determination of the 50% inhibition concentration (IC 50 ) and inhibition constant ( K i ) of each novel phloroglucinol derivatives, three concentrations of each derivative were tested. The activities (%) were plotted against the concentrations in Lineweaver–Burk plots, as previously described . The IC 50 and K i values were calculated by curve fitting the data.…”
