Secondary metabolites produced by Fusarium sporotrichioides DAOM 165006 in liquid culture

Abstract: A Canadian strain of Fusarium sporotrichioides, DAOM 16005, grown in liquid culture produced a variety of trichothecenes. The yields obtained in two different fermentation media, MYRO and Moss and Franks, were approximately the same; however, in the later medium, T-2 formed more rapidly and then was degraded to HT-2. All the trichothecenes isolated were 3,4-oxygenated compounds; T-2 was the major metabolite together with neosolaniol, HT-2, and DAS. Of some 16 minor metabolites detected, four were characterized… Show more

“…The 1D 1 H NMR data is consistent with and complements previous NMR findings on T-2 toxin [42,43,44,45]. In the current experiment, the chemical shift and coupling constants of the hydrogens attached directly to the multiple ring skeleton confirms the previously defined stereochemistry.…”

“…Most notably, the cross peaks between hydrogens: H-7 β and H-13 A , H-7 α and H-14, H-2 and H-13 B , H-13 A and H-14, H-15 A and H-14, H-15 AB and H-4, support the assignments made by Greenhalgh et al [43,44,45]. Prominent EXSY crosspeaks were observed between water and H-3 OH (Figure 4), indicating the presence of chemical exchange, which was further confirmed with the deuterium exchange experiment (Figure 2B).…”

“…In the present work, the two major trichothecenes produced by F. poae were DAS and FX. Although type-B trichothecenes were produced in small amounts by F. sporotrichioides, the major trichothecenes were the A-type, as previously reported (Scott et al 1980, Ueno & Ishii 1985, Greenhalgh et al 1988.…”

“…Infected kernels with roots were dried at 42 °C in a thermostat and ground into flour in a laboratory mill. To study the influence of PME on in vitro DON production by F. culmorum , the fungus was gown in a submerged culture at 220 rpm and 25 °C for 7 days on toxinogenesis-promoting liquid Myro medium [73,74] supplemented with the extract to the final concentrations of 0.5 or 1.0 mg/mL. The fungus culture from PME-free medium obtained under the same conditions served as a control.…”
