Aims: Several fungal endophytes were isolated from some medicinal plants and screened for their ability to produce the anticancer drug vinblastine. Methods and Results: An isolate was found to produce vinblastine (205Á38 μg l −1 ), and the identity of the fungal vinblastine was confirmed by UV spectroscopic, high-performance liquid chromatography and electrospray ionization mass spectrometry analyses. Based on both morphological and molecular studies, the vinblastine-producing strain was identified as Alternaria alternata. Cytotoxic activities of the fungal vinblastine were evaluated against CHO-K1, MCF-7 and HepG-2 cell lines by the MTT assay. The proliferation of these cell lines was inhibited after treatment with fungal vinblastine and the recorded IC 50 values of the respective cell lines were 12Á15, 8Á55 and 7Á48 μg ml −1 . A strain improvement programme for improving vinblastine productivity by the fungal strain was also used. In addition, 10 broth media were evaluated for further increasing the production of vinblastine. The yield of vinblastine was intensified by 3Á98-fold following gamma irradiation at 1000 Gy, and a stable mutant strain was isolated. Among the screened media, M1D broth (pH 6Á0) stimulated the highest vinblastine production of 1553Á62 μg l −1 by the isolated mutant strain. Conclusions: The present study is the first report on the production and yield improvement of the anticancer drug vinblastine by A. alternata. Significance and Impact of the Study: These findings suggest A. alternata as a viable and potent source with excellent biotechnological potential for the production of vinblastine.

Alternaria spp. cause different diseases in potato and tomato crops. Early blight caused by Alternaria solani and brown spot caused by Alternaria alternata are most common, but the disease complex is far more diverse. We first provide an overview of the Alternaria species infecting the two host plants to alleviate some of the confusion that arises from the taxonomic rearrangements in this fungal genus. Highlighting the diversity of Alternaria fungi on both solanaceous hosts, we review studies investigating the genetic diversity and genomes, before we present recent advances from studies elucidating host–pathogen interactions and fungicide resistances.TaxonomyKingdom Fungi, Phylum Ascomycota, Class Dothideomycetes, Order Pleosporales, Family Pleosporaceae, Genus Alternaria.Biology and host rangeAlternaria spp. adopt diverse lifestyles. We specifically review Alternaria spp. that cause disease in the two solanaceous crops potato (Solanum tuberosum) and tomato (Solanum lycopersicum). They are necrotrophic pathogens with no known sexual stage, despite some signatures of recombination.Disease symptomsSymptoms of the early blight/brown spot disease complex include foliar lesions that first present as brown spots, depending on the species with characteristic concentric rings, which eventually lead to severe defoliation and considerable yield loss.ControlGood field hygiene can keep the disease pressure low. Some potato and tomato cultivars show differences in susceptibility, but there are no fully resistant varieties known. Therefore, the main control mechanism is treatment with fungicides.

Effect of Alternaria alternata (Fr.) Keissl. on the physiology of Pea (Pisum sativum L.)

Alternaria alternata (Fr.) Keissl. is responsible for causing leaf spots and blight diseases on a large number of horticultural and agricultural crops including pea. The response and resistance of different pea varieties to this fungus might vary. Therefore, this study was aimed to investigate the varietal differences in the physiological response of Pisum sativum L. to A. alternata. Various physiological and biochemical parameters of two commercial pea varieties (Leina and Meature) were assessed at vegetative stage in pot experiment. Sterilized seeds were soaked in 1×105 mL conidial suspension of A. alternata for 15 min and control seeds were soaked in sterilized water. These sterilized seeds were sown in pots containing sterilized soil. In both varieties, germination percentage, germination rate, seedling vigor index, root and shoot length, dry and fresh biomass, leaf sugar, chlorophyll a, b, total chlorophyll and carotenoid contents, leaf osmotic potential, leaf relative water content, significantly decreased under A. alternata stress as compared to control. But proline and protein content significantly increased. It is concluded from the results that A. alternata caused adverse effects on the physiology of pea plant. The tested pea variety Leina was found to be more sensitive under fungal stress of A. alternata as compared to Meature and the extent of stress caused by both test varieties can be exploited in future studies.
