Second to fourth digit ratio: a predictor of adult penile length

Abstract: The second to fourth digit ratio (2D:4D) has been proposed as a putative biomarker for prenatal testosterone and covaries with the sensitivity of the androgen receptor (AR). Both prenatal testosterone and the AR play a central role in penile growth. In this study, we investigated the relationship between digit ratio and penile length. Korean men who were hospitalized for urological surgery at a single tertiary academic centre were examined in this study, and 144 men aged 20 years or older who gave informed con… Show more

“…To minimize measurement errors, the mean values of two digit ratios calculated from the duplicate measurements were used in the present analysis. The intraclass correlation coeffi cient of two repeated measures of the digit ratio by a single investigator was 0.935, which was similar to that reported in previous studies (intraclass correlation coeffi cient = 0.873 -0.969) [ 16,18 ] . This means that direct measurement of the digit ratio using a digital vernier caliper has acceptable repeatability [ 19,20 ] .…”

“…Figure 1 illustrates the method for measuring digit length. As described in our previous two studies [ 15,16 ] , before PSA level determination and TRUS, right-hand second and fourth digit lengths were measured by a single investigator using a digital vernier caliper accurate to an accuracy of 0.01 mm, directly on the ventral surface of the fi ngers, from the crease proximal to the palm at the base of each digit to the digit tip [ 17 ] ( Fig. 1 ).…”

“…The Homeobox genes, Hoxa and Hoxd, regulate the differentiation of urogenital system, and may have an effect on the prenatal testicular androgen synthesis and digit development (Kondo et al, 1997;Peichel et al, 1997). Moreover, Choi et al (2011) suggested that digit ratio can predict adult penile size and that the effects of prenatal T may in part explain the difference in adult penile length. A high 2D : 4D ratio in right hands of males was associated with germ cell failure due to azoospermia and oligospermia (Manning et al, 2000).…”

“…Thus, digit ratio is primarily determined during embryonic development1516 and changes little after sexual maturation 1617. Recently, it has been reported that digit ratio was associated with testosterone levels in infertile men18 and the risk of prostate cancer19 and penile length 2021…”
