The S-glycoprotein (Spike) of the SARS-CoV-2 forms a complex with the human transmembrane protein angiotensin-converting enzyme 2 (ACE2) during infection. It forms the first line of contact with the human cell. The FDA-approved drugs and phytochemicals from Indian medicinal plants were explored. Molecular docking and simulations of these molecules targeting the ACE2–Spike complex were performed. Rutin DAB10 and Swertiapuniside were obtained as the top-scored drugs as per the docking protocol. The MD simulations of ligand-free, Rutin DAB10-bound, and Swertiapuniside-bound ACE2–Spike complex revealed abrogation of the hydrogen bonding network between the two proteins. The principal component and dynamic cross-correlation analysis pointed out conformational changes in both the proteins unique to the ligand-bound systems. The interface residues, His34, and Lys353 from ACE2 and Arg403, and Tyr495 from the Spike protein formed significant strong interactions with the ligand molecules, inferring the inhibition of ACE2–Spike complex. Few novel interactions specific to Rutin-DAB10 and Swertiapuniside were also identified. The conformational flexibility of the drug-binding pocket was captured using the RMSD-based clustering of the ligand-free simulations. Ensemble docking was performed wherein the FDA-approved database and phytochemical dataset were docked on each of the cluster representatives of the ACE2–Spike. The phytochemicals identified belonged to Withania somnifera , Swertia chirayita , Tinospora cordifolia and Rutin DAB10, fulvestrant, elbasvir from FDA. Introduction:Kuṅkumādi Ghṛta (KG) is an Ayurvedic formulation commonly recommended in skin disorders, especially Mukhadūṣikā (Acne vulgaris). Kesara (stigma of Crocus sativus Linn.) is a prime ingredient in the formulation. However, due to high cost and increased adulteration in Kesara, ‘Nāgakesara’ (Mesua ferrea Linn.) is suggested by Ayurvedic experts as a substitute. Nāgakesara is relatively lower in cost and possesses similar therapeutic attributes to that of Kesara. Recent studies have established standard manufacturing procedures and clinical efficacies of Kesarayukta (with Kesara) Kuṅkumādi Ghṛta (KKG) and Nāgakesara yukta (with Nāgakesara) Kuṅkumādi Ghṛta (NKG) in Mukhadūṣikā. In spite of wide utility of KG in Ayurvedic practice, no published work on its shelf life is available so far.Aims and Objectives:To establish the physicochemical profile and evaluate the shelf life of KKG and NKG.Materials and Methods:Total three batches of KKG and NKG each were prepared in the laboratory by adopting Ayurvedic classical guidelines and the findings were systematically recorded. Comparative differences in organoleptic characters (colour, odour, taste, touch) and values of different physicochemical parameters (Ash value, pH value, Total solid content, Specific gravity, Refractive index, Loss on drying, Acid value, Iodine value, Saponification value, Peroxide value and Rancidity tests) of raw materials [herbal ingredients, Go Ghṛta, Mūrcchita Ghṛta (MG)] as well as finished products (KKG and NKG) were recorded. Long term (real time) shelf life testing was carried out at the intervals i.e., at 0, 3, 6, 9 and 16 months. High performance thin layer chromatography (HPTLC) analysis of samples was also carried out.Results and Conclusion:The average percentage loss in final product was 10.18% in KKG and 7.58% in NKG. The shelf life of MG, KKG and NKG is 16 months. After 16 months, Go Ghṛta showed negative rancidity test, while MG, KKG and NKG showed positive rancidity, indicative of onset of deterioration after 16 months. Present findings validate the Ayurvedic principles stipulating medicated Ghṛta to have a shelf life of 16 months. Chromatographic study showed few differences between KKG and NKG samples. Data obtained by present study may be considered as standard for future researches.

Tinospora cordifolia is a requisite medicinal plant, has been used for the treatment of various diseases and has been recommended for improving the immune system mentioned in Ayurveda and other systems of medicine as a monoherbal or polyherbal preparation. Nonetheless, Ayurvedic and other ancient literature reported that Tinospora cordifolia growing on neem plant gives better immunomodulatory activity. The traditional and scientific data comprising immunomodulatory potential, pharmacognostic description, therapeutic uses, phytochemical constitution, of Tinospora cordifolia and neem guduchi using the aqueous extract in the form of satwa has well established. The present study evaluates the comparative immunomodulatory activity using satwa of Tinospora cordifolia and neem guduchi using in-vitro models, for understanding the influence of edaphic factors on biologically active compounds. Evaluation of the immuno-modulatory potential of the guduchi satwa and neem guduchi satwa at the dose of 150 and 300 mg/kg body weight was done in rats; assessment of immunomodulatory activity was carried out by assessment of neutrophil adhesion whereas phytochemical profile was established by HPTLC method. Simultaneously, the correlation between soil factors and medicinally active compound responsible for immunomodulation also studied. In the present study, we have compiled and investigated the active compounds which are immunomodulator. Results of present studies suggested that neem guduchi satwa was found to be potent immuno-stimulant in a dose-dependent manner when compared with other group and there is no clear correlation between soil factors and medicinally active compound were observed.
