“…The TJ proteins claudin-1, claudin-4, and occludin were detected as dots and whisker-like lines in the superficial layers. The TJ strand is reported to be restricted to locations where occludin is present although the function of occludin remains unclear (9,35,36,39). Therefore, we focused on claudins colocalized with occludin at junctions to analyze the functions of TJ in epithelial layers.…”

“…7 However, more conclusive evidence of epidermal TJs in human adult skin was not established until the beginning of the millennium. [8][9][10] Almost at the same time, the notion of a functional TJ barrier in mammalian epidermis was revived by the observation that epidermal permeability barrier dysfunction developed in claudin (cldn)-1 knockout and cldn-6 transgenic mice. 11,12 Unlike the simple monolayer epithelium present in the gastrointestinal tract and bronchi where each cell expresses the full range of cellular junctions (e.g., adherens junctions, desmosomes, gap junctions, TJs), the epidermis is a highly stratified structure where cells from different layers express unique combinations of junctional complexes.…”

“…Accordingly, TJ-related structures have been identified by EM below and above the SG, especially in the interdesmosomal region of various stratified epithelia. 9,17 Over the past 2 decades outstanding progress has been made in this field, but much remains to be done to fully appreciate the role epidermal TJs play in human health and disease. In this paper we will review what is known about the epidermal TJ barrier function and composition, its regulation by the immune system as well as the relevance of these observations for dermatologic diseases.…”

“…9 The TJ strand has been said to be restricted to wherever occludin exists, even if the function of occludin is still unclear. [23][24][25][26][27] Even in the stomach and intestine, the expression pattern of the claudins is broad, running from the apical to basolateral membrane, while that of occludin is only dense at TJs in normal tissues. 12,28 Figure 5 Influence of acid on TJ protein localization in ALI cells.…”

“…This is consistent with the data from skin. 24 As the functional TJ proteins are insoluble in nonionic detergent, 10,12,29,30 in the present study, proteins from ALIcultured cells were divided into nonionic detergent (NP-40)-soluble and -insoluble fractions, and the level of claudin-1 and claudin-4 in each fraction then measured. Claudin-4 levels were lower in pH 2-stimulated cells compared with control conditions, in a time-dependent manner, suggesting that acid stimulation delocalized the claudin-4 from the TJs in the superficial layers.…”
