Scribble downregulation in adenomyosis compromises endometrial stromal decidualization by decreasing FOXO1 expression

Abstract: STUDY QUESTION Does Scribble (SCRIB) contribute to aberrant decidualization of endometrial stromal cells (ESC) in adenomyosis? SUMMARY ANSWER SCRIB knockdown impairs decidualization of ESC by decreasing Fork-head box O1A (FOXO1) expression through the protein kinase B (AKT) and atypical protein kinase C (aPKC) activated pathways. WHAT IS KNOWN ALREADY Stromal… Show more

“…The expression of LIF and HOXA11, which are important for embryo implantation, 19–31 is disturbed in the endometria of patients with adenomyosis 32 . Furthermore, patients with adenomyosis exhibit impaired decidualization due to the reduced expression of HOXA10, FOXO1, KLF5, CEBPB, HAND2, 33 and SCRIB, 34 which are crucial for decidualization, as shown in mouse experiments 35–39 . A recent study suggested the involvement of somatic KRAS mutations in the pathogenesis of adenomyosis 40 .…”

“…The expression of ITGB3 increases in the mid-secretory phase, which corresponds to WOI. Hence, ITGB3 is also regarded as a decidualization marker in ESCs [ 61 ]. In our study, we established a mechanism where the protein and mRNA levels of HOXA10 and ITGB3 increased significantly in ESCs treated with 0.1 IU/mL hCG for 72 h. This finding implies that hCG may promote decidualization and enhance endometrial receptivity by increasing the expression levels of HOXA10 and ITGB3 in ESCs.…”

“…Adenomyosis is characterized by pain (dysmenorrhoea, dyspareunia, and chronic pelvic pain), AUB, and subfertility. The mechanism by which adenomyosis causes subfertility is not fully understood, but women with adenomyosis show impaired decidualization ( Jiang et al , 2016 ), including reduced expression of Scribble ( Peng et al , 2021 ). The underlying mechanisms resulting in AUB are equally ill-defined.…”
