Plants have evolved different defense mechanisms to combat pathogen attacks. In this study, detached leaf assays were conducted to estimate the influence of leaf position (leaf age) on the development of chocolate spot disease, caused by Botrytis fabae, in the lower, middle and upper leaves of five faba bean (FB) cultivars with different levels of resistance/susceptibility. Two components of resistance in terms of lesion diameter and spores per lesion were used to evaluate the disease intensity. To evaluate plant defense response, oxidative burst and phenol-oxidizing enzymes were assayed. The results indicated that regardless of the resistance level of the FB cultivar, lower (older) leaves were more severely infected by the chocolate spot pathogen than upper (younger) ones. A comparison of the defense-response behavior of the FB leaves revealed that the chocolate spot pathogen induced lipid peroxidation and the production of reactive oxygen species, peroxidase, and polyphenol oxidase in leaf tissue during the FB-B. fabae interaction. The production of these defense compounds in leaves was not static but governed in time and extent by physiological maturity. Younger leaves exhibited significantly higher oxidizing enzyme activity and lower oxidative stress than older ones. These variations in the levels of defense compounds could explain the differences in leaf resistance. The extreme differences in disease development recorded in upper and lower leaves of all FB cultivars suggest that assessing resistance using leaves from the middle positions would be the most efficient and reliable for evaluating the resistance/susceptibility of FB plants to B. fabae.

QTL analysis for ascochyta blight resistance in an intraspecific population of chickpea (Cicer arietinum L.)

In both controlled environment and the field, six QTLs for ascochyta blight resistance were identified in three regions of the genome of an intraspecific population of chickpea using the IDS and AUDPC disease scoring systems. One QTL-region was detected from both environments, whereas the other two regions were detected from each environment. All the QTL-regions were significantly associated with ascochyta blight resistance using either of the disease scoring systems. The QTLs were verified by multiple interval mapping, and a two-QTL genetic model with considerable epistasis was established for both environments. The major QTLs generally showed additive gene action, as well as dominance inter-locus interaction in the multiple genetic model. All the QTLs were mapped near a RGA marker. The major QTLs were located on LG III, which was mapped with five different types of RGA markers. A CLRR-RGA marker and a STMS marker flanked QTL 6 for controlled environment resistance at 0.06 and 0.04 cM, respectively. Other STMS markers flanked QTL 1 for field resistance at a 5.6 cM interval. After validation, these flanking markers may be used in marker-assisted selection to breed for elite chickpea cultivars with durable resistance to ascochyta blight. The tight linkage of RGA markers to the major QTL on LG III will allow map-based cloning of the underlying resistance genes.

Necrotrophic pathogens of the cool season food legumes (pea, lentil, chickpea, faba bean and lupin) cause wide spread disease and severe crop losses throughout the world. Environmental conditions play an important role in the development and spread of these diseases. Form of inoculum, inoculum concentration and physiological plant growth stage all affect the degree of infection and the amount of crop loss. Measures to control these diseases have relied on identification of resistant germplasm and development of resistant varieties through screening in the field and in controlled environments. Procedures for screening and scoring germplasm and breeding lines for resistance have lacked uniformity among the various programs worldwide. However, this review highlights the most consistent screening and scoring procedures that are simple to use and provide reliable results. Sources of resistance to the major necrotrophic fungi are summarized for each of the cool season food legumes. Marker-assisted selection is underway for Ascochyta blight of pea, lentil and chickpea, and Phomopsis blight of lupin. Other measures such as fungicidal control and cultural control are also reviewed. The emerging genomic information on the model legume, Medicago truncatula, which has various degrees of genetic synteny with the cool season food legumes, has promise for identification of closely linked markers for resistance genes and possibly for eventual map-based cloning of resistance genes. Durable resistance to the necrotrophic pathogens is a common goal of cool season food legume breeders.
