Screening of hyaluronic acid–poly(ethylene glycol) composite hydrogels to support intervertebral disc cell biosynthesis using artificial neural network analysis

Abstract: Hyaluronic acid (HA) poly(ethylene glycol) (PEG) composite hydrogels have been widely studied for both cell delivery and soft tissue regeneration applications. A very broad range of physical and biological properties have been engineered into HA-PEG hydrogels that may differentially affect cellular “outcomes” of survival, synthesis and metabolism. The objective of this study was to rapidly screen multiple HA-PEG composite hydrogel formulations for an effect on matrix synthesis and behaviors of nucleus pulposus… Show more

“…A study of the influence of the functional group ratio (defined as the number of thiol groups over the number of vinyl sulfone groups, SH/VS) on particle size and the crosslinking www.nature.com/scientificreports www.nature.com/scientificreports/ occurrence, is conducted. Literature data on the SH/VS ratio are reporting a value of 1.2 and a total polymer concentration of 6% w/V as the optimized condition for batch processes [61][62][63] .…”

“…Some groups have designed biocompatible scaffolds that may incorporate features supportive of the NP cell phenotype, such as the inclusion of native extracellular matrix components, such as hyaluronan, proteoglycans, or collagens (102, 103) (104). In one example, a polymerizing fibrin containing the growth factor transforming growth factor β (TGF-β) was delivered with allogeneic mesenchymal stem cells and shown to preserve disc height and to inhibit apoptosis beyond that of either the growth factor or cells alone (105).…”

“…The Barricaid is under current clinical investigation with promising results including decreased disability, increased IVD height and reduced rate of recurrent IVD herniation, but had a higher frequency of endplate complications at 2) and 3 years . In addition to mechanical repairs, there are a range of AF replacement materials including electrospun poly‐e‐caprolactone and hyaluronic acid with polyethylene (material properties reviewed in Reference ). Void filling biomaterials which can be injected with or without cells, such as collagen, were recently reviewed in Reference .…”
