Screening of a Novel Fibrinolytic Enzyme-Producing Streptomyces from a Hyper-Arid Area and Optimization of Its Fibrinolytic Enzyme Production

Abstract: Fibrinolytic enzymes are a kind of proteolytic enzymes that can hydrolyze fibrin and dissolve blood clots. They could be used as a therapeutic agent for treating thrombosis. It is important for the treatment of cardiovascular disease to find and develop new thrombolytic drugs. In order to explore new fibrinolytic enzymes, a strain named 214L-11 with protease and fibrinolytic enzyme activity, which was isolated from the Flaming Mountain of Xinjiang Province, was screened using the skimmed milk plate, the blood … Show more

“…The composition and concentration of the medium can exert a significant influence on microbial fermentation production [45][46][47]. This is particularly applicable to microbial secondary metabolic processes, where the composition and concentration of the carbon sources in the medium can significantly impact the fermentation of secondary metabolites, such as pigment fermentation and antibiotic production [48][49][50]. The choice of carbon source can exert a significant influence on the biosynthesis of specific secondary metabolites, owing to its capacity to inhibit gene expression and repress enzyme activity [51].…”

“…The CNXK100 protease has been found to possess fibrinolytic activity, a characteristic documented in a few other studies involving proteases derived from S. radiopugnans VITSD8 ( Verma et al 2018 ), S. rubiginosus VITPSS1 ( Dhamodharan et al 2019 ), and Streptomyces sp. 214L–11 ( He et al 2023 ). Due to its stability towards pepsin, trypsin, chymotrypsin, and fibrinolytic activity, the CNXK100 protease holds potential applications in medications ( Bose et al 2019 ).…”

“…This enzyme bears resemblance to the one initially isolated from the Gobi radiation zone by our research group [ 31 ]. Undoubtedly, abnormal environments, such as the ocean’s high salinity, high pressure, and polar environment, offer promising prospects for future microbiology and biotechnology studies [ 32 ].…”
