Objectives: Tuberculosis (TB) remains a global public health problem. Recent studies have implicated long non-coding RNAs and their variants as possibly playing important roles in TB. The aim of this study was to assess the clinical relevance of lnc-AC145676.2.1-6 and lnc-TGS1-1 and their variants in a western Chinese population. Methods: This case-control study included 467 TB patients and 473 healthy controls from West China Hospital. The expression levels of lnc-AC145676.2.1-6 and lnc-TGS1-1 were analyzed by reverse transcriptase quantitative real-time PCR. Single-nucleotide polymorphism genotyping was performed using a custom-designed 2 Â 48-Plex SNPscan kit. Results: It was observed that lnc-AC145676.2.1-6 and lnc-TGS1-1 expression levels were both obviously down-regulated in TB patients. In addition, a lower expression level of lnc-TGS1-1 was associated with the presence of thrombocytopenia in TB patients during anti-TB treatment, and the homozygous CC genotype of rs4737420 correlated with a decreased risk of leukopenia, compared with individuals with the T allele (TT/CT genotype), in the dominant mode. Conclusions: For the first time, potential TB-associated promoting effects were identified for the decreased expression levels of lnc-AC145676.2.1-6 and lnc-TGS-1, while lnc-TGS1-1 and its variant rs4737420 may be predictive indicators of anti-TB drug-induced adverse drug reactions. Larger validation studies on different populations are warranted to confirm these findings.

A tuberculose é uma infecção micobacteriana comumente causada pelo agente etiológico Mycobacterium tuberculosis, podendo também ser desencadeada por outras bactérias do gênero Mycobacterium. Ainda que bem documentada, mais de um século após sua descoberta, situa-se como a doença infecciosa que mais mata no mundo. Configura-se também problema socioeconômico importante, atingindo em especial as camadas mais pobres da população e comprometendo seriamente sua capacidade produtiva. Os métodos diagnósticos tradicionais para a doença consistem da baciloscopia e cultura, recentemente adotando os testes moleculares rápidos, todos com limitações significativas, como a incapacidade de discriminar entre casos de tuberculose ativa e latente, sensibilidade inconsistente e incapacidade de oferecer prognósticos. A presente revisão da literatura procurou elucidar os atuais avanços na pesquisa de biomarcadores da infecção nos últimos 5 anos com o emprego da metodologia ELISA. Foram verificadas e comparadas evidências apresentadas pelos estudos e as performances diagnósticas alcançadas em cada contexto. Constatou-se uma evidente heterogeneidade nas respostas imunológicas entre populações e indivíduos, provável fruto da diversidade genética do patógeno que possui linhagens intercontinentais e regionais, bem como diferenças particulares nas expressões de alguns biomarcadores, seja por variáveis genéticas ou mesmo outras doenças de base; a homologia dos antígenos pesquisados com outras espécies de bactérias também foi uma problemática. Fica claro que questões genéticas particulares de populações e do próprio agente infeccioso precisam ser melhor consideradas nesses estudos, a fim de alcançar um perfil sorológico suficientemente sensível e específico para triagem clínica e diagnóstico destes pacientes.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (Mtb), is one of leading causes of global deaths. This study aimed to explore the role of miR‐18a in RAW264.7 cells response to Mtb infection. Exosomes derived from Mtb‐infected cells were isolated and further validated by size, transmission electron microscopy and Western blot. RT‐PCR was utilized to measure miR‐18a expression. Cell viability and ultrastructure were examined by CFU counting, CCK‐8 and electron microscope, respectively. Potential target genes of miR‐18a were predicted with bioinformatics and further confirmed using RT‐PCR, Western blot and laser confocal microscope analysis, respectively. LC3, AMPK and mTOR were measured using Western blot. We found that miR‐18a was induced both in Mtb‐infected RAW264.7 cells and its derived exosomes compared with the controls. In addition, up‐regulation of miR‐18a promoted intracellular Mtb survival, attenuated cell viability and reduced LC3‐II level, while its down‐regulation had the opposite effect. miR‐18a overexpression suppressed level of ATM, one possible target of miR‐18a, while its underexpression enhanced ATM. We also found that inhibition of ATM induced LC3‐II decrease in Mtb‐infected cells and could reverse the increase of LC3‐II caused by inhibition of miR‐18a. Moreover, down‐regulation of miR‐18a increased p‐AMPK level while reduction of ATM could reverse the change. Taken together, our results suggest that miR‐18a is up‐regulated in macrophages response to Mtb infection, and it promotes intracellular Mtb survival through repressing autophagic process by down‐regulation of ATM pathway. This provides new thought for TB pathogenesis, diagnosis and treatment.
