Screening and genetic analysis of resistance to peanut stunt virus in soybean: identification of the putative Rpsv1 resistance gene

Abstract: The peanut stunt virus (PSV) causes yield losses in soybean and reduced seed quality due to seed mottling. The objectives of this study were to determine the phenotypic reactions of soybean germplasms to inoculation with two PSV isolates (PSV-K, PSV-T), the inheritance of PSV resistance in soybean cultivars, and the locus of the PSV resistance gene. We investigated the PSV resistance of 132 soybean cultivars to both PSV isolates; of these, 73 cultivars exhibited resistance to both PSV isolates. Three resistant… Show more

“…Peanut stunt virus (PSV) is a pathogen of plants, especially legumes, which belongs to genus Cucumovirus [ 1 , 2 , 3 , 4 , 5 ]. The PSV genome consists of three (+)single-stranded RNAs and five open reading frames (ORFs) encoding the 1a and 2a components of the viral replicase, the movement protein, the coat protein (CP), and the 2b protein known as a viral silencing suppressor [ 6 ].…”

“…SMV inoculation was carried out according to Saruta et al (2012) . SMV-infected soybean leaf tissue frozen at –80°C was homogenized in 0.1 M sodium phosphate buffer (pH 7.0) using a ratio of approximately 1 g of infected tissue per 10 mL of buffer.…”

“…Sugimoto et al (2008) mapped Rps1d to chromosome 3 using progenies from a cross between 'Tanbakuro' and 'PI 103091' to determine selectable SSR markers (Sugimoto et al 2008). PSV resistance is traced to a single dominant gene, Rpsv1, near Satt435 on chromosome 7 by using RILs with PSV resistance from a cross between 'Hyuga' and 'Enrei' (Saruta et al 2011). Two QTLs for antibiosis resistance to CCW, CCW -1, and CCW -2, both on chromosome 7, have been identified using progenies from a cross between 'Fukuyutaka' and 'Himeshirazu' (Komatsu et al 2005).…”
