YY1 binds to the E3ʹ enhancer and inhibits the expression of the immunoglobulin κ gene via epigenetic modifications

Abstract: The rearrangement and expression of immunoglobulin genes are regulated by enhancers and their binding transcriptional factors that activate or suppress the activities of the enhancers. The immunoglobulin κ (Igκ) gene locus has three important enhancers: the intrinsic enhancer (Ei), 3' enhancer (E3'), and distal enhancer (Ed). Ei and E3' are both required for Igκ gene rearrangement during early stages of B-cell development, whereas optimal expression of the rearranged Igκ gene relies on both E3' and Ed. The tra… Show more

“…YY1 is ubiquitously expressed in human tissues and controls various cellular mechanisms including transcriptional regulation (activation/repression), cell proliferation, DNA repair, chromatin modeling, apoptosis, autophagy, X-chromosome inactivation, recruitment of Polycomb Group (PcG) proteins and epigenetic modifications (1, 2, 1013). YY1 has also been implicated in B cell maturation, development and immunoglobin class switch recombination (14, 15).…”

“…Given that pro‐inflammatory phenotype of adventitial fibroblasts from the PAH vessel are usually associated with epigenetic alterations (Gomez‐Arroyo et al, 2016; Lau et al, 2017), and YY‐1 acts predominantly as an epigenetic modulator (Infante et al, 2015; Zhou et al, 2018), we subsequently examined the alterations of histone methylation modifications of pulmonary adventitial fibroblasts by western blotting. As shown in Figure 3A, sCD40L treatment did not obviously affect the modificatory levels of H3K4me3 and H3K36me3, which are commonly considered to activate gene transcription (Guenther et al, 2007; Edmunds et al, 2008).…”
