Gastric cancer (GC), the second most common malignant cancer worldwide, gives rise to a number of cancer-associated fatalities annually. Accumulating evidence has shown that microRNAs (miRs) may serve as oncogenes or tumor suppressors in GC development. The aim of this study was to discover the expression, function and mechanism of miR-761 in GC progression. First, the findings revealed that the expression level of miR-761 was significantly decreased in GC cell lines and tissues. The functional studies showed that miR-761 in GC cells inhibited tumor proliferation and metastasis. In the mechanistic study, through an online database search and luciferase assay, Ras and Rab interactor 1 (RIN1), which has been demonstrated as an oncogene in various types of cancer, including GC, was identified as a target of miR-761. Notably, miR-761 expression was demonstrated to be negatively correlated with RIN1 mRNA levels in GC tissues. Simultaneously, overexpression of RIN1 partially rescued the inhibitory effect of miR-761 mimic in the GC cells. The present study provided new insights into the role of miR-761 in the progression of GC, and implicated the potential application of miR-761 in GC cell therapy.

Retinoblastoma (RB) is the most common intraocular malignancy in childhood, and the prognosis in the advanced RB is poor. It is urgent to find novel therapeutic targets. Long noncoding RNAs (lncRNAs) have critical functions in cancer progression, and lncRNA LINC00202 is found associated with poor prognosis in RB. However, the functions of LINC00202 in RB remain unclear. We employed qRT-PCR and immunoblot to detect the expression levels of mRNAs and proteins, respectively. Cell proliferation was determined by CCK-8 assay and colony formation assay. Transwell assays were applied to evaluate the cell abilities of migration and invasion. Luciferase reporter assay was applied to examine RNA stability, and RNA pulldown assays were used to detect interaction between lncRNA and microRNA (miRNA). LINC00202 expression in RB tissues is higher than that in the paired adjacent normal tissues, which has correlation with poor prognosis in RB. RB cell proliferation, migration and invasion were weakened by LINC00202 depletion, but enhanced by LINC00202 overexpression. MiR-3619-5p was identified to directly bind and mediate LINC00202-promoted RB progression, meanwhile, miR-3619-5p directly regulated expression of an oncongene, RIN1. Moreover, RIN1 knockdown completely blocked miR-3619-5p-enhanced RB progression. In summary, high LINC00202 levels are correlated with poor prognosis in RB, and it promotes RB progression by sponging miR-3619-5p and therefore up-regulating RIN1 expression.

Renal cell carcinoma (RCC) is one of the most common urological malignancies occurring in adult human kidneys worldwide. Recent research on antitumor drugs has focused on plant extracts, a class of compounds that play critical roles in cancer treatment. The present study aimed to investigate the potential antitumor effect of ginkgolic acid (GA) in RCC. Transwell invasion assay, cell counting kit-8 assay and flow cytometry were used to measure cell migration, cell viability and apoptosis, respectively. A network pharmacology approach was applied to identify pathway information, combining molecular docking techniques to screen for key target information. In the present study, GA inhibited the viability and proliferation of RCC cells (786-O and A498), both in vitro and in vivo, via G 1 arrest. GA also reduced RCC cell invasion and migration. In addition, the epidermal growth factor receptor (EGFR) was identified as a critical target protein of GA, which significantly inactivated EGFR signaling in RCC (P<0.05). Collectively, the present study provided evidence that GA exerts its anticancer function by directly targeting the EGFR signaling pathway, revealing the potential of GA therapy for RCC.
