II. Yeast sequencing reports. The sequence of a 22·4 kb DNA fragment from the left arm of yeast chromosome II reveals homologues to bacterial proline synthetase and murine α‐adaptin, as well as a new permease and a DNA‐binding protein

Abstract: We report the sequencing of a 22,470 bp DNA fragment from the left arm of Saccharomyces cerevisiae chromosome II. Thirteen open reading frames longer than 300 bp provisionally called YBL0520, YBL0401 to YBL0408 and YBL0410 to YBL0413 have been detected. Five genes were previously sequenced: COR1, encoding a core protein of the mitochondrial coenzyme QH2 cytochrome c reductase complex (Tzagaloff and Crivellone, 1986), PRS3, a proteasome subunit gene (Lee et al., 1992), ERD2, coding for a protein involved in the… Show more

“…The clustering between COG0325 genes and proC observed decades ago (De Wergifosse et al, 1994) was confirmed; clustering between yggS and proC was observed in *20 % of the genomes analysed (3 % in the 1000 genome set; Table 1). However, our data showed that the link between yggS and proline synthesis is not robust (Fig.…”

“…1). This observation gave the PROline synthase co-transcribed homolog (PROSC) name to the first studied members of the family including the human homolog, which is expressed in all organs (De Wergifosse et al, 1994). Finally, in yeast, the COG0325 protein YBL036C was induced threefold in response to the DNA-damaging agent methyl methanesulfonate (MMS), implying involvement in processes ensuring genetic integrity (Lee et al, 2007).…”

“…An open reading frame for an L16-like protein was found in the flanking DNA of the published sequence of the S.cerevisiae URA7 gene (13,21 Figure 2 indicates that Rmll6p is located in the mitochondrial fraction (lane labeled 'mt') and co-sediments specifically with the 54S large ribosomal subunit. To test whether Rml16p is an essential protein in the mitochondrial ribosome, an nnlJ6A::URA3 gene disruption allele (Fig.…”

“…Bacterial PROSC may be involved in proline synthesis, and it may be co-transcribed with proC, which is a known proline biosynthetic gene 13) , and its predicted amino acid sequence was highly homologous to the product encoded by a Caenorhabditis elegans (C. elegans) gene, F09.0814 14) . Therefore, this gene is a biologically interesting gene and is likely to play a vital role in cellular function.…”
