Morus nigra L. extract prolongs survival of rats with hepatocellular carcinoma

Abstract: Morus nigra is a rich source of anthocyanins, phytochemicals that have anticancer effects. This study aimed to investigate the effects of M. nigra extract (MNE) on diethylnitrosamine (DEN)-induced hepatocellular carcinoma (HCC). Male Sprague-Dawley rats were assigned into four groups (n = 10): control, DEN, and DEN +100 or 400 mg/kg of MNE. After 4 months, the DEN group showed a significant mortality rate, hepatic lipid peroxidation, dysplastic nodules in the cirrhotic liver, and an increase of blood bilirubin,… Show more

“…However, DEN signi cantly increased the expression of Akt, Wnt, and β-catenin and signi cantly decreased the expression of PTEN. These ndings are in agreement with our previous work that DEN changed the expression of PTEN, Wnt, and β-catenin while had no signi cant effect on the PI3K gene [34]. Treatment with R. turkestanicum extract (400 mg/kg) could restore the PTEN expression and downregulated the expression of Wnt4 and β-catenin genes.…”

“…Also, carcinogenesis by DEN is strongly associated with the excessive production of free radicals and decreasing the anti-oxidant enzymes which trigger time-dependent necrosis through accumulating oxidized thiol proteins [30,31]. In recent years, studies have reported bene cial effects of natural compounds in the prevention and management of cancers including HCC [32][33][34]. It has been shown the extract has anti-oxidant activity and anti-cancer effects against different cancer cell lines [11].…”

“…In DEN/PB+LMFP and DEN/PB+HMFP groups, rats were administrated exactly as DEN/PB group and additionally orally administered a low dose of MFP (LMFP) at 50 mg/kg body weight or HMFP at 100 mg/kg body weight daily for 14 successive weeks. The choice of MFP dose and treatment time was based on previous literature [ 20 , 28 , 29 ]. The detailed timelines were shown in Figure 1a .…”

“…To date, only limited mouse and rat studies have shown a reduction in WNT4 expression in HCC models. 28,29 WNT4 is a secretory ligand, and its function has been reported in wound healing and angiogenesis. 30 The level of WNT4 varies among different cancer types.…”
