DNA identification of species of the Anopheles maculipennis complex and first record of An. daciae in Belgium

Abstract: The present study aimed at identifying the members of the Anopheles maculipennis complex (Diptera: Culicidae) occurring in Belgium. Therefore, the second internal transcribed spacer of nuclear ribosomal DNA (ITS2) and the mitochondrial cytochrome oxidase subunit I (COI) loci were sequenced in 175 and 111 specimens, respectively, collected between 2007 and 2019. In parallel, the suitability of two species‐diagnostic PCR‐RFLP assays was tested. The identified specimens included: An. maculipennis s.s. (N = 105), … Show more

“…Morphological identification of adults and larvae was done with a stereomicroscope using dichotomic and digital keys [ 29 , 32 37 ]. To confirm and validate the morphological identification of IMS and to identify damaged adults and larvae, DNA-barcoding was performed [ 18 , 26 , 27 , 36 , 38 , 39 ].…”

However, other authors consider this polymorphism to be an indicator of different forms/ecotypes that are consistent with the different geographical origins of the tested populations [ 32 , 88 , 115 117 ]. On the other hand, other authors have analyzed genomic differences and chromosomal inversions, especially on the X -chromosome, between An.…”

“…Anopheles daciae as a distinct species was identified first in Romania, then described using a molecular approach in Germany [ 41 44 , 119 122 ], England and Wales [ 56 , 123 ], the Czech Republic and Slovakia [ 39 ], Serbia [ 108 ] and more recently in Poland [ 124 ], Finland [ 101 ], Sweden [ 116 ] and Russia [ 18 ]. It has also been found in Italy and Belgium, with the provisional definition of Anopheles daciae species inquirenda [ 32 , 117 ].…”
