Traumatic brain injury (TBI) survivors face debilitating long-term psychosocial consequences, including social isolation and depression. Acute TBI modifies neurovascular physiology and behavior but a gap in our understanding are the chronic physiological implications of altered brain perfusion on behavioral activities, particularly social interactions.We investigated longitudinal functional vascular networks across the brain for 2-months post- TBI and its impact on social behavior. Adult C57/BL6 male mice received a moderate cortical TBI. Behavior (foot-fault, open-field, 3-chamber social preference) was assessed at baseline, 3-, 7-, 14-, 30-, and 60-days post injury (dpi) followed by magnetic resonance imaging (MRI, 9.4T). Anatomical MRI (T2-weighted), dynamic susceptibility contrast (DSC) perfusion weighted MRI (PWI) were acquired at each temporal epoch. After the final 60dpi MRI, animals underwent transcardial perfusion fixation to map angioarchitecture. MRI data were analyzed using standardized protocols followed by cross-correlations between social behavior, cerebral perfusion, and vascular metrics.Social behavior deficits at 60dpi emerged as reduced interaction with a familiar cage-mate (partner). We observed multiphasic decrements in cerebral blood flow (CBF) encompassing lesion and perilesional cortex where acute reductions at 3-14dpi partially recovered by 30dpi, followed by significant reductions in perfusion at 60dpi. The CBF perturbations extended antero-posteriorly from the ipsilateral TBI impact site but also adulterated contralateral brain regions. CBF reductions impacted regions known to regulate social behavior including hippocampus, hypothalamus, and rhinal cortex. Alongside perfusion deficits at 60dpi, social isolation in TBI-mice emerged with a significant decline in preference to spend time with a cage mate. Cortical vascular density was also reduced corroborating the decline in brain perfusion and social interaction.Thus, the novel temporal neurovascular loss, and subsequent recovery followed by chronic decrements are broadly reflected by social interaction perturbations. Our correlations strongly implicate a linkage between vascular density, cerebral perfusion, and social interactions, where early evaluation can potentially predict long-term outcomes. Thus, our study provides a clinically relevant timeline of alterations in functional vascular recovery that can guide research for future therapeutics.

The benefits of sociality bring a hidden burden as constant social interactions increase the probability of pathogen transmission. When facing diseases, simple strategies such as social distancing have proven effective in preventing disease spread. Cleaning interactions are textbook examples of multispecies sociality, where cleaner organisms remove ectoparasites, diseased and injured tissue from a heterospecific client. These interactions can be potential vectors of infectious diseases in reef fishes, being characterised as potential disease hotspots. Here, we aimed to determine if, in the context of infections, cleaner wrasses (Labroides dimidiatus) employ social distancing and other behavioural tactics to prevent disease spread within the context of cleaning interactions. To this end, we used bacterial lipopolysaccharides (LPS) immunostimulation to induce sickness behaviour in cleaner wrasses without harming the fish. After LPS immunostimulation, we performed four behavioural trials: the social preference test (from cleaner and client perspectives), the social interaction test, and the bystander cooperativeness test. In the social preference test, LPS-injected cleaners showed lower activity rates but no change in their social preference. However, two tested client species preferred sick cleaners. Contrarily, during the social interaction test, cleaners reduced the number of interactions, despite clients' willingness to interact. For the bystander test, LPS-injected cleaners were unable to adjust their behaviour to be more cooperative. Reduced activity levels revealed lethargy, which may lead to passive self-isolation, allowing for some social distancing of healthy individuals. The findings of these experiments suggest that while feeling sick, immune-stimulated cleaners undergo changes in their behaviour which can be viewed as a passive social distancing-like barrier preventing disease spread. This is especially relevant as some clients prefer and continue to request interactions from sick cleaners.

Path integration is a strategy that allows animals to monitor their movement relative to a starting point using directional and distance cues. It has been observed in a wide range of terrestrial species, but evidence of this behaviour in fish is still lacking. In contrast to most animals shown to navigate via path integration, fish are not surface bound but exist within a three-dimensional medium. This may present additional challenges when monitoring their position relative to a specific point. We developed a novel experimental paradigm to test whether the coral reef-dwelling domino damselfishDascyllus trimaculatuscould use path integration to navigate back to a shelter location. This consisted of a circular pool with a shelter and landmarks near the edge and a trap containing food in the centre. Our results show that these fish follow homing trajectories back to the previous location of the removed shelter without using landmarks after a foraging trip into the central trap. Error in homing trajectory increased with outward path length, consistent with the use of path integration. These results addD. trimaculatusto the growing list of path-integrating species, opening new avenues for investigating the evolution and neural underpinnings of this navigational strategy.
