Scianna antigens including Rd are expressed by ERMAP

Abstract: The Scianna blood group encompasses the high-frequency antigens Sc1 and Sc3 and the low-frequency antigen Sc2. Another lowfrequency antigen Rd (Radin) was suggested to belong to the Scianna blood group. The molecular basis of the Scianna blood group was unknown. The erythrocyte membrane-associated protein (ERMAP) shared the genomic location, protein product size, and localization to the red blood cell (RBC) membrane surface with Scianna. The ERMAP gene was sequenced in probands with known Scianna and Radin phe… Show more

“…The Sc2 allele is a consequence of the ERMAP(Gly57Arg) polymorphism found at a frequency of approximately 1% in Northern Europeans. Similarly, Radin is a consequence of ERMAP(Pro60Ala), a rarer allele (125).…”

“…Artificial DNA fragments were generated for the following LFAs and their antithetical HFA partners: SC2 (SC*02), LW [17][18][19][20][21]. Before use, artificial DNA fragments were titrated to equimolar copy concentrations as found in genomic DNA extracts from donor samples before validation.…”

“…These include Rh-ced (which encodes an allelic Rh antigen complex), Ermap (which encodes the Scianna blood group antigen 63,64 ), Duffy, and ICAM4 (also known as the LW antigen 65 ; Table 3). These genes were validated as EKLF target genes in the fetal liver by real-time PCR (Figures 1 and 3), although only Duffy and Ermap were significantly upregulated by tamoxifen in B1.6 cells ( Figure 1D).…”
