Carotenoids are widely distributed and structurally diverse, which have significant roles in the photosynthesis of plants. As a precursor of vitamin A, carotenoids are also antioxidants that reduce various chronic diseases, which are beneficial for human health. Currently, the existing studies concerned the biological roles of APETALA2 (AP2)/ethylene-responsive factor (ERF) genes originated from higher plants. The AP2 superfamily of the transcriptional regulator was identified in higher plants, which was related to growth, development, carotenoid metabolism, and responses to various stresses. However, the regulatory mechanisms of the AP2-modulating carotenoid metabolism have not been reported in microalgae, which remain to be elucidated. Dunaliella parva AP2 (i.e., DpAP2), an important transcription factor, promotes carotenoid accumulation by binding to the promoter of target gene. Here, we identified an important AP2/ERF transcription factor, DpAP2, which could promote carotenoid accumulation by binding to the promoter of target gene. To demonstrate the function of DpAP2, the interacting proteins were identified by the yeast two-hybrid system. The results showed that DpAP2 could interact with three proteins with different activities (DNA-binding transcription factor activity, protein kinase activity, and alpha-D-phosphohexomutase activity); these proteins may be associated with multiple biological processes. This paper laid a good foundation for a deep understanding of the regulatory mechanisms of DpAP2 and genetic engineering breeding in D. parva.

The two major bottlenecks faced during microalgal biofuel production are, (a) higher medium cost for algal cultivation, and (b) cost-intensive and time consuming oil extraction techniques. In an effort to address these issues in the large scale set-ups, this comprehensive review article has been systematically designed and drafted to critically analyze the recent scientific reports that demonstrate the feasibility of microalgae cultivation using wastewaters in outdoor raceway ponds in the first part of the manuscript. The second part describes the possibility of bio-crude oil production directly from wet algal biomass, bypassing the energy intensive and time consuming processes like dewatering, drying and solvents utilization for biodiesel production. It is already known that microalgal drying can alone account for ∼30% of the total production costs of algal biomass to biodiesel. Therefore, this article focuses on bio-crude oil production using the hydrothermal liquefaction (HTL) process that converts the wet microalgal biomass directly to bio-crude in a rapid time period. The main product of the process, i.e., bio-crude oil comprises of C16-C20 hydrocarbons with a reported yield of 50–65 (wt%). Besides elucidating the unique advantages of the HTL technique for the large scale biomass processing, this review article also highlights the major challenges of HTL process such as update, and purification of HTL derived bio-crude oil with special emphasis on deoxygenation, and denitrogenation problems. This state of art review article is a pragmatic analysis of several published reports related to algal crude-oil production using HTL technique and a guide towards a new approach through collaboration of industrial wastewater bioremediation with rapid one-step bio-crude oil production from chlorophycean microalgae.

Algal lipids are considered to be good raw materials towards aqua feed formulation and biofuels production, and hence, they have been investigated for decades. Using external carbon source, microalgal cultivation has been getting attention due to high biomass productivity and lipid content with flexible cultivation of algae. In this present study, a marine microalga, Tetraselmis suecica was cultivated by combined method with sodium acetate as carbon source and reciprocal nutrients concentration. The reciprocal concentration of nitrogen and phosphorus with sodium acetate supply towards lipid and fatty acid methyl esters (FAME) synthesis in terms of live feed efficiency was attempted. The highest FAME content (78.44 ± 1.18% of DW) of alga was obtained in nil nitrogen condition with limited phosphate concentration (N0‐PL) followed by N0‐P (77.31 ± 2.5% of DW) combination. Limitation of phosphate with combination of nil nitrogen (N0‐PL) showed slight increment of total FAME accumulation and its higher increments resulted in mono‐unsaturated fatty acids (MUFA) accumulation, indicating nitrogen starvation was a real FAME trigger under heterotrophic cultivation. Effect of heterotrophic cultivated T. suecica feeding on marine copepod Pseudodiaptomus annandalei produced higher population density (119 ± 15 ind. L−1) in N0‐P combination and high rate (71.98%) of nauplii production was obtained in N0‐PL combination. These findings suggested that the heterotrophic microalgae cultivation can be practiced for aqua feed (especially for copepod cultivations) formulation as well as to phosphorous removal from wastewaters.
