Scaled-expansion of the membrane associated cytoskeleton requires conserved kinesin adaptors

Abstract: A periodic lattice of actin rings and spectrin tetramers scaffolds the axonal membrane. How spectrin is delivered to this structure to scale its size to that of the growing axon is unknown. We found that endogenous spectrin, visualized with singe axon resolution in vivo, is delivered to hotspots in the lattice that support its expansion at rates set by axon stretch-growth. Unlike other cytoskeletal proteins, whose apparent slow movement consists of intermittent bouts of fast movements, spectrin moves slowly an… Show more

“…Briefly, the injection mix contained 0.25 μg/μl S.pyogenes Cas9 (IDT), 0.1 μg/μl tracrRNA (IDT), 0.056 μg/μl crRNA, 25 ng/μl pre-melted double-stranded DNA repair template and 40 ng/μl PRF4 (rol-6 (su1006)) plasmid. The mix was injected into a strain that expresses GFP1-10 fragment in the DA9 neuron and another neuron in the ventral cord 72 . Non-roller F1 progeny from P0 roller jackpot plates were screened by single worm PCR.…”
