Scalable biomimetic sensing system with membrane receptor dual-monolayer probe and graphene transistor arrays

Abstract: Affinity-based biosensing can enable point-of-care diagnostics and continuous health monitoring, which commonly follows bottom-up approaches and is inherently constrained by bioprobes’ intrinsic properties, batch-to-batch consistency, and stability in biofluids. We present a biomimetic top-down platform to circumvent such difficulties by combining a “dual-monolayer” biorecognition construct with graphene-based field-effect-transistor arrays. The construct adopts redesigned water-soluble membrane receptors as s… Show more

“…Notably, the regulatory functions of stromal cell-derived factor-1 (SDF-1) and its receptor chemokine receptors C-X-C chemokine receptor type 4 (CXCR4) are crucial in governing the survival and differentiation processes of stem cells ( Janssens et al, 2018 ). The SDF-1/CXCR4 signaling pathway is implicated in the modulation of cellular developmental and migratory activities ( Qing et al, 2023 ). Additionally, it plays a significant role in the facilitation of nerve regeneration and repair mechanisms.…”

“…The independence of available structure inputs and complex computational programs makes QTY code promising to be a simple method of water-soluble protein design. The initial application of QTY code was on chemokine receptors (a class of G-protein coupling receptors, GPCRs), exhibiting favorable water-solubility, stability, and ligand-binding affinity 12 , 14 , and showing excellent sensitivity and robustness in a biomimetic sensing system 15 . Nonetheless, to what extent QTY code can preserve the biological activities needs to be further investigated.…”
