Scalable agroinfiltration-based production of SARS-CoV-2 antigens for use in diagnostic assays and subunit vaccines

Abstract: Agroinfiltration is a method used in biopharming to support plant-based biosynthesis of therapeutic proteins such as antibodies and viral antigens involved in vaccines. Major advantages of generating proteins in plants is the low cost, massive scalability and the rapid yield of the technology. Herein, we report the agroinfiltration-based production of glycosylated SARS-CoV-2 Spike receptor-binding domain (RBD) protein. We show that it exhibits high-affinity binding to the SARS-CoV-2 receptor angiotensin-conver… Show more

“…The described methodology was adapted from the surrogate neutralization ELISA assay, as shown in Colwill et al and Demone et al, for the evaluation of the relative inhibition of neutralizing antibodies to spike or RBD protein from binding to soluble ACE2 [ 57 , 58 ]. Briefly, SARS-CoV-2 spike or RBD protein were diluted in sterile 1X PBS to 8 μg/mL and coated onto 384-well Immuno plates (12.5 μL/well) overnight at 4 °C.…”

“…The RBD alone and its fusion with the Fc region of human IgG1 (RBD-Fc) are utilized as an antigen to develop subunit vaccines. The recombinant RBD and RBD-Fc showed specific binding to human ACE2 receptor [ 51 , 70 , 71 , 72 ]. In animal tests, the plant-produced RBD and RBD-Fc antigens elicited potent neutralizing responses in mice and non-human primates [ 51 , 70 ].…”
