Scaffolds Loaded with Dialdehyde Chitosan and Collagen—Their Physico-Chemical Properties and Biological Assessment

Abstract: In this work, dialdehyde chitosan (DAC) and collagen (Coll) scaffolds have been prepared and their physico-chemical properties have been evaluated. Their structural properties were studied by Fourier Transform Infrared Spectroscopy with Attenuated Internal Reflection (FTIR–ATR) accompanied by evaluation of thermal stability, porosity, density, moisture content and microstructure by Scanning Electron Microscopy—SEM. Additionally, cutaneous assessment using human epidermal keratinocytes (NHEK), dermal fibroblast… Show more

“…We assessed the impacts of solvents of ellagic acid (80HA/20EA, 50HA/50EA, and 20HA/80EA) on the viability of cutaneous cells (human dermal fibroblasts and epidermal keratinocytes) compared with human melanoma models (A375 and G361) ( Figure 3 ). The selection of cell lines was in accordance with previous reports [ 25 , 26 , 27 ]. The obtained results demonstrated the significant decrease in the proliferation ratio versus the control cells, in both healthy and tumor cells.…”

“…The material’s byproducts and unreacted chemicals cannot be removed after repeated washing [ 52 ]. Reactants such as 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide (EDC) and N-hydroxysuccinimide (NHS), genipin, and dialdehyde starch (DAS), which are significantly less toxic to cells, are substitutes for glutaraldehyde [ 53 , 54 , 55 ]. EDC-NHS is a “zero-length” crosslinking agent that can facilitate the formation of amide bonds by linking the carboxyl group of glutamic or aspartic acid collagen to the amino group.…”

“…Ряд авторов также проводил исследования хитозан-коллагеновых каркасов ex vivo и in vivo, в ходе которых была выявлена высокая степень неоваскуляризации формирующихся на каркасе тканей, а также ускорение эпителизации раны [1,4,5]. Помимо повышенной эпителизации, в моделях ран было также продемонстрировано увеличение скорости пролиферации и повышение клеточной адгезии кератиноцитов in vitro [2,7,8].…”
