Sasa veitchii extracts suppress acetaminophen-induced hepatotoxicity in mice

Abstract: Background: The aim of this study was to investigate the therapeutic effects of a Sasa veitchii leaf extract (SE) on acetaminophen (APAP)-induced hepatotoxicity. Methods: Seven-week-old male ddY mice were orally administered SE or saline (0.2 mL) once a day for a week. Twenty-four hours after the last pretreatment, the mice were intraperitoneally injected with 550 mg/kg APAP or saline under fasting conditions. The mice from each group were euthanized and bled for plasma analysis 2, 6, 24, and 72 h after the in… Show more

“…Plasma levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were determined using a Transaminase CII kit (Wako Pure Chemical Industries, Osaka, Japan), as previously described [ 24 26 ]. Plasma levels of tumor necrosis factor alpha (TNFα) were measured using a commercially available ELISA kit (eBioscience, San Diego, CA, USA), as previously described [ 22 ]. For relative quantification, calibration curves were prepared using standard solutions.…”

“…For the detection of nuclear DNA strand breaks, paraffin-embedded sections were stained with the terminal dUTP nick-end labeling (TUNEL) stain using an in situ apoptosis detection kit (Takara Bio, Shiga, Japan) according to the manufacturer's protocols. The sections were counterstained with hematoxylin (23). For immunohistochemical analysis, the paraffin-embedded sections were deparaffinized and rehydrated in a graded ethanol series.…”

“…In accordance with other extracts from natural products, 7 days of a Sasa veitchii (Carrière) Rehder leaf extract (0.2 ml per day) pretreatment significantly decreased hepatic injury markers, including ALT and AST, oxidative stresses, including malondialdehyde and glutathione, histologic damages, inflammatory cytokines, activation of JNK, receptor-interacting protein-1, which has emerged as a key modulator of necrotic cell death, and CYP2E1, whereas the total antioxidant capacity within the liver was increased in an acute APAP mice model (550 mg/kg) (Zhang et al, 2014;Yoshioka et al, 2017). More recently, the seed and bark extracts (400 mg/kg) of Acacia catechu (L.f.) Willd.…”
