Sargachromenol Purified from Sargassum horneri Inhibits Inflammatory Responses via Activation of Nrf2/HO-1 Signaling in LPS-Stimulated Macrophages

Abstract: In this study, we isolated sargachromenol (SC) from Sargassum horneri and evaluated its anti-inflammatory effect in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophages. SC did not show cytotoxicity at all concentrations and effectively increased the cell viability by reducing the nitric oxide (NO) and intracellular reactive oxygen species (ROS) production in LPS-stimulated RAW 264.7 macrophages. In addition, SC decreased the mRNA expression levels of inflammatory cytokines (IL-1β, IL-6, and TNF-α) and in… Show more

“…Ethanol, methanol, and ethyl acetate S. serratifolium extracts, whose major compounds identified were the meroterpenoids SHQA, SC, and SQA, were reported to dose-dependently (1.25–10 µg/mL) reduce LPS-induced ROS in LPS-stimulated macrophages [ 56 ]. The same trend was observed with SCM and SHQA isolated from S. horneri and S. macrocarpum , respectively [ 48 , 49 ]. In turn, an acetone:ethanol extract of S. glaucescens rich in fucoxanthin caused a decrease in LPS-induced ROS and O 2 •− production in the same cell line at concentrations ranging from 25 µg/mL to 100 µg/mL [ 122 ].…”

“…These results agree with those found in the literature [ 50 , 51 ], where brown seaweeds also revealed anti-inflammatory potential. Han and co-workers [ 52 ] evaluated the anti-inflammatory effect of sargacromenol isolated from Sargassum horneri in RAW 264.7 macrophages stimulated by LPS. Sargacromenol, besides reducing the production of NO and ROS, also decreased the mRNA expression of inflammatory cytokines, namely IL-1β, IL-6 and TNF-α, and inflammatory mediators, namely iNOS and COX-2.…”

“…Nuclear magnetic resonance (NMR) spectroscopy was conducted to elucidation of Sargachromenol structure (400 MHz, JNM-ECX400, JEOL, Japan). 1H (CD3OD, 400 MHz) and 13C (CD3OD, 100 MHz) spectra were obtained [36] (Figure 5).…”
