Sample preparation followed by HPLC under harmless 100% aqueous conditions for determination of oxytetracycline in milk and eggs

Abstract: A simple and hazardous chemical-free method for the high-performance liquid chromatographic determination of oxytetracycline (OTC) residues in milk and eggs has been developed. Sample preparation consists in homogenization with an aqueous solution by means of a handheld ultrasonic homogenizer followed by centrifugal ultrafiltration. HPLC is performed with an isocratic aqueous mobile phase and a photodiode array detector. Average recoveries of OTC (0.05, 0.1, and 0.2 microg mL(-1) for milk; 0.1, 0.2, and 0.4 mi… Show more

“…The centrifugal ultrafiltration procedure is easy to use and requires little time and solvent, but usually results in high recovery and reproducibility [12,13]. Samples containing NOV were prepared by methanol extraction of chicken muscle followed by a standard centrifugal ultrafiltration procedure.…”

“…This procedure allows a minimal consumption of serum sample (about 100 lL), is simple, rapid and easily applicable in a routine analysis laboratory providing a satisfactory clean up. Ultrafiltration has been extensively used for the isolation and enrichment of proteins [37,38] and for clinical diagnosis of several diseases [36]; very few applications as clean-up tool have been reported [39][40][41][42].…”

“…Antibiotics of the TC series are usually determined by microbiological approaches [5], immunoassays [6], HPLC [7][8][9][10], and CE [11][12][13][14]. Microbiological approaches and immunoassays are expensive and time-consuming with low sensitivity and poor specificity.…”
