Sample Collection and Preparation of Biofluids and Extracts for Liquid Chromatography-Mass Spectrometry

Abstract: Nowadays, metabonomics has been widely applied to the area of biomedicine. Based on metabolic profiling analysis, metabolomic studies can provide information of the metabolic phenotype affected by genetic or environmental factors. Liquid chromatography-mass spectrometry has been proven to be a robust platform for metabolic profiling by sensitive measurement of low molecular weight compounds. However, this sensitive platform requires standard protocols in the preanalytical stage to avoid unwanted results caused… Show more

“…Fifty milligrams of freeze-dried liver power were weighed for each sample (placed in ice water until the next step). Metabolite extraction was carried out using 1.5 mL of methanol/water solution (methanol:water = 4:1, vol:vol) in the presence of a 5 mm diameter stainless steel bead using a homogenizer (Spex Sample PREP, Stanmore, UK) operating at 20 Hz for 2 min and stood for 60 min at −20 °C ( Yin et al., 2015 ). The mixture was then centrifuged at 30,000 × g for 3 min at 4 °C.…”
