Salt stress is an environmental signal affecting degradative enzyme synthesis in Bacillus subtilis

Abstract: Growth under conditions of salt stress has important effects on the synthesis of degradative enzymes in Bacillus subtilis. Salt stress strongly stimulates the expression of sacB, encoding levansucrase (about ninefold), and downregulates the expression of aprE, encoding alkaline protease (about sixfold). It is suggested that the DegS-DegU two-component system is involved in sensing salt stress. Moreover, it has been shown that the level of sacB expression strongly depends on the growth conditions; its expressio… Show more

“…B. subtilis was transformed with plasmid or chromosomal DNA according to the two-step protocol described previously (Kunst and Rapoport, 1995). Transformants were selected on SP plates containing chloramphenicol (Cm, 5 g ml ¹1 ), kanamycin (Km, 5 g ml ¹1 ), tetracycline (Tc, 15 g ml ¹1 ), spectinomycin (Spc, 100 g ml ¹1 ), phleomycin (Phl, 0.2 g ml ¹1 ), or erythromycin plus lincomycin (Em 1 g ml ¹1 and Lin 10 g ml ¹1 , respectively).…”

“…This fragment was ligated to pMutin-4 previously digested with the same enzymes. The resulting plasmid, pCL29, was used to transform B. subtilis YB886 competent cells prepared as indicated by Kunst & Rapoport (1995). Since the amplicon contains an internal segment of the ywiE gene, Erm R colonies raised by homologous recombination led to a knockout mutation.…”
