Salt concentrations in culture media for the development of Dipteryx alata in vitro

Abstract: -The objective of this work was to evaluate the effect of salt concentrations in culture media on the early development of "barueiro" (Dipteryx alata) seedlings, obtained from the shoot apex, in vitro. Two different in vitro culture media -Murashige & Skoog (MS) and wood plant medium (WPM) -in different salt concentrations (25, 50, 75, and 100%) were used. At 120 days, the D. alata seedlings in the MS medium, in its original concentration (100%), developed better than in other concentrations or in the WPM. The… Show more

“…The delay in germination of H. coddii subsp. barnardii seeds cultured on full and 1¼ strength of MS media could be ascribed to the higher salt concentration in MS medium as compared to other culture media with a lower salt content used for in vitro seed germination (Araruna et al, 2017;Davies et al, 2017). Solanki and Siwach (2012) observed no germination when seeds of Aconitum heterophyllum were cultured on cotton wool moistened with MS medium or on solid MS medium, and seeds only germinated on cotton wool moistened with distilled water.…”

“…can be attributed to the lower salt concentrations (especially nitrogen and potassium) (Tomasi et al, 2018). For the in vitro establishment of sprouting apexes of Dipteryx alata Vog., the MS medium was considered to be more adequate compared to the WPM, providing seedlings with longer root length and higher leaf number, and an increase in dry mass was observed with the salt concentrations used (25; 50; 75, and 100%) for both in vitro culture media (Araruna et al, 2017).…”

“…Nesse sentido, a cultura de tecidos constitui um avanço para obtenção de materiais propagativos, em quantidade de mudas certificadas e qualidade de padrão fitossanitário e genético das mesmas. Todavia, ainda são escassos relatos envolvendo a cultura in vitro de Dipteryx alata (MAMEDES & ARAÚJO, 2010, SILVA et al, 2016ab, ARARUNA et al, 2017REZENDE et al, 2019).…”
