Salmonella typhimurium LT2 possesses three distinct 23S rRNA intervening sequences

Abstract: The rrl genes for 23S rRNA of Salmonella typhimurium LT2 are known to carry intervening sequences (IVSs) at two sites, helix-25 and helix-45, which are excised by RNase III during rRNA maturation, resulting in rRNA which is fragmented but nevertheless functional. We isolated DNA fragments containing the seven rrl genes from BlnI, I-CeuI, and SpeI genomic digests following pulsed-field gel electrophoresis and used these DNA fragments as templates for PCRs utilizing primers upstream and downstream of helix-25 an… Show more

“…Two specific tig transcripts were detected in the tig + strains that were absent in the tig deletion mutant. The larger transcript was detected near the 16S rRNA (∼1.5 kb in size; used as loading control) (Mattatall and Sanderson 1996). The other tig transcript was smaller in size.…”

“…Cellular total RNA used for deep sequencing (RNA-seq) was isolated using the hot phenol method (Mattatall and Sanderson 1996;Beckmann et al 2010). Cellular total RNA for Northern blotting and primer extension was prepared using the TRIzol (Ambion) method.…”

“…However, while the overall 16S rRNA gene sequence similarity between Bacillus strain 171544 T and clone BPC094 is 95n7 %, the nucleotide similarity of the insertion sequences in helix 49 is only 49 % indicating that if they do have a common origin, these insertions are subject to rapid evolutionary change. It has not been demonstrated whether the insertion is present in the 16S rRNA molecule or removed by processing, as it is for example in Clostridium paradoxum (Rainey et al, 1996) and Salmonella typhimurium (Mattatall & Sanderson, 1996), because of difficulties obtaining reverse transcriptase products from purified rRNA (data not shown).…”
