Salmonella enterica serovar Choleraesuis infection of the porcine jejunal Peyer's patch rapidly induces IL-1β and IL-8 expression

Abstract: Salmonella enterica serovar Choleraesuis is an enteric pathogen of swine, producing septicemia, enterocolitis, pneumonia, and hepatitis. The initial molecular events at the site of Salmonella infection are hypothesized to be critical in the initiation of innate and adaptive immune responses; however the acute immune response elicited by porcine intestinal tissues is not well understood. To address this need, we employed explants of jejunal Peyer's patch (JPP) mucosa from pigs to examine Salmonella-induced immu… Show more

“…The cDNA was diluted 5-fold, before equal amounts were added to duplicate qRT-PCR reactions. Tested genes of interest were IL-8 [22], IL-6 [23], TNF-α [24] and COX-2 [19]. Hypoxanthine phosphoribosyltransferase (HPRT) [25] and Cyclophilin-A (CycA) [24] were used as reference genes.…”

“…Various animal and tissue models have been established in swine for investigation of mucosal immune mechanisms and induction of protective memory. They include respiratory models of bacterial and viral infection (Baarsch et al, 2000; Baarsch et al, 1995; Saif, 1996; Saif et al, 1994; VanCott et al, 1994), enteric viral infection and defined antigen models (Brim et al, 1995; Foss and Murtaugh, 1999a, 2000; Hyland et al, 2004; Hyland et al, 2006b; Lanza et al, 1995; Saif, 1999; VanCott et al, 1993), ex vivo secretory tissue preparations in Ussing chambers, and epithelial transwell cell culture systems (Gookin et al, 2004; Hyland et al, 2006a; Schmidt et al, 2007). …”

“…Among more than 2500 Salmonella serotypes, S. enterica serotype Choleraesuis has a high predilection for causing systemic infection in swine, including enterocolitis, septicemia, and pneumonia (Hyland, Brown, & Murtaugh, 2006), and in humans, including bacteremia and mycotic aneurysm (Chen et al, 2007;Jean, Wang, & Hsueh, 2006). Antimicrobial therapy is inevitable for such patients (Cohen, Bartlett, & Corey, 1987;Su & Chiu, 2007).…”
