SAGA–CORE subunit Spt7 is required for correct Ubp8 localization, chromatin association and deubiquitinase activity

Abstract: Background Histone H2B deubiquitination is performed by numerous deubiquitinases in eukaryotic cells including Ubp8, the catalytic subunit of the tetrameric deubiquitination module (DUBm: Ubp8; Sus1; Sgf11; Sgf73) of the Spt-Ada-Gcn5 acetyltransferase (SAGA). Ubp8 is linked to the rest of SAGA through Sgf73 and is activated by the adaptors Sus1 and Sgf11. It is unknown if DUBm/Ubp8 might also work in a SAGA-independent manner. Results … Show more

“…observed in the ada1Δ and spt7Δ mutant strains. This observation is consistent with the fact that Ada1 and Spt7 belong to the CORE of SAGA and their absence has previously been shown to fully impair SAGA complex integrity(Nuño-Cabanes et al, 2020;Sterner et al, 1999;Wu & Winston, 2002). Consistently, Ada1 and Spt7 recruitment at NCR-sensitive genes were fully interdependent.…”

“…Ada1 also functions in delimiting the boundaries of gene silencing in chromatin and this function was linked to proteasome processes ( Kamata et al, 2016 ). In addition, Ada1 is also involved in repairing radiation damage ( Game et al, 2005 ), regulation of gene expression in the yeast metabolic cycle ( Sánchez-Gaya et al, 2018 ), and nuclear localization of Ubp8 subunit ( Nuño-Cabanes et al, 2020 ).…”
