The aim of the present study was directed to investigate the therapeutic efficacy of cetuximab and curcumin either in combination or alone as anticancer treatment mmodality on 7,12-dimethylbenz[a]anthracene (DMBA) induced hamster buccal pouch(s) (HBP(s)) carcinoma. Material and methods: Twenty-Five Syrian male hamsters, five weeks old, weighing 80-120g were divided into three group(s) (G(s)): A, B, and C. G-A (negative control): HBP mucosa of 5 animals were painted with liquid paraffin alone three times a week for 18 weeks. G-B: 20 right HBP animals were painted with 0.5% DMBA in paraffin oil 3 times a week for 18 weeks. Then animals were divided randomly into four subgroups. G-B1 (DMBA treated group): 5 animals were left untreated for 6 weeks after DMBA painting. G-B2 (cetuximab treated group): 5 animals were injected intraperitoneally with cetuximab 1 mg/animal at 3 days intervals for 6 weeks following DMBA-treatment. G-B3 (curcumin treated group): Right HBP of 5 animals were directly injected with curcumin 80 mg/kg body weight every day for 6 weeks following DMBA-treatment. G-B4 (curcumincetuximab treated group): 5 animals were directly injected with curcumin followed by intraperitoneal injection of cetuximab for 6 weeks following a DMBA-treatment. The assessment was based on the gross observation, histological and immunohistochemical examination utilizing anti-apoptotic Bcl-2 antibody. Results: Gross observation revealed variation in reduction of the tumor size in the treated groups (G-B2, G-B3, and G-B4) compared to that observed in group G-B1. Histopathological findings revealed variations among the treated groups. Immunohistochemical results revealed that Bcl-2 expression has a variability in the area % throughout the groups used. Groups G-B1, G-B2, G-B3 and G-B4 were 70.7 %, 54.8 %, 42.7% and 24.5% respectively. Conclusion: The synergistic effect of both cetuximab and curcumin resulted in promising therapeutic effect in regression of DMBA induced HBP carcinoma.

The aim of the present study was directed to screen the oncogenic Kras gene mutation in 7,12-dimethylbenz[a]anthracene (DMBA) induced hamster buccal pouch(s) (HBP(s)) carcinoma. Material and methods: Ten Syrian male hamsters, five weeks old, weighing 80-120g were divided into two group(s) (G(s)). GI (negative control): Right HBP mucosa of 5 animals were painted with liquid paraffin alone three times a week for 18 weeks. GII: (DMBA treated group): Right HBP of 5 animals were painted with 0.5% DMBA in paraffin oil 3 times a week for 18 weeks. After termination of the experiment, the HBPs were excised and bisected with one section fixed in 10% neutral buffered formalin, routinely processed and embedded in paraffin blocks in order to be sectioned and examined histologically. The other sections of the fresh tissues were taken and stored at-80°C for DNA extraction followed by amplification of the targeted sequence using the polymerase chain reaction (PCR) technique. Then, the amplified targeted sequences were sequenced using the chain termination method (Sanger method) with a small concentration of dideoxynucleotides to detect the possible presence of Kras gene mutation. Results: Histological sections, using H&E stain, showed moderately to poorly differentiated SCC. DNA sequencing results revealed that Kras gene mutation was absent in all cases. Conclusion: Oncogenic activation of Kras gene is not important in DMBA induced HBP carcinoma.

Backgrounds: Oral squamous cell carcinoma (OSCC) is among the most frequent oral cancers in individuals under 40. Documents have endorsed that a diet enriched with fruit and vegetables can banish the risk of developing major cancers. This study aimed to evaluate the effects of different concentrations of four medicinal herbs including saffron, ginger, cinnamon and curcumin on OSCC cell line. Methods: Having obtained the aqueous extract of the four herbs, they were administered on OSCC cell lines per se and in dual, triple, and quadruple combinations. Their effects were measured in different concentrations and in 24 and 48 hours by using MTT assay. Results: The minimum and maximum effective concentrations were respectively 108 and 217 mg/ml for curcumin with IC30 of 77mg/ml, 108 and 270 mg/ml for ginger with IC30 of 58 mg/ml, 2 and 10 mg/ml for saffron with IC30 of 1.9 mg/ml, and 5 and 40 mg/ml for cinnamon with IC30 of 3.3 mg/ml. The best effect of the combinations was seen for cinnamon-saffron after both 24 and 48 hours and the four herbs combination after 48 hours. Conclusion: Although all the four herbs were effective on OSCC cell line, the strongest extract was saffron, followed by cinnamon. Combination of cinnamon-saffron and combination of the four herbs showed maximum effects. These findings suggest that traditional medicinal herbs may potentially contribute to oral cancer treatment; providing new windows for the development of new therapeutic strategies for OSCC.
