Safety of the standardized quality tree sublingual immunotherapy tablet: Pooled safety analysis of clinical trials

Abstract: Background The standardized quality (SQ) tree sublingual immunotherapy (SLIT)‐tablet has recently been approved for treatment of tree pollen allergy. Healthcare workers should be provided with detailed safety data for clinical use. Objective To assess the tolerability and safety of the SQ tree SLIT‐tablet (12 SQ‐Bet) in adults and adolescents. Methods Safety data were pooled from three double‐blinded, randomized, placebo‐controlled trials (2 phase‐II/1 phase‐III) including adults and adolescents 12–65 years wi… Show more

“…Forty‐three percent of the participants in the LNIT group experienced local reactions, such as sneezing, mild rhinorrhea, nasal congestion, itching, and a local burning sensation. This rate was comparable with local reactions caused by SLIT (39%−47%) 52,53 . Nickelsen et al 30,31 and Cirla et al 40 reported a high incidence of local AEs.…”

“…This rate was comparable with local reactions caused by SLIT (39%−47%). 52,53 Nickelsen et al 30,31 and Cirla et al 40 reported a high incidence of local AEs. This high incidence could be attributable to the variation of the report degree accepted by each article without the same report standard.…”

“…These extracts are manufactured from one or more natural allergen sources including pollens, arthropods (mites), animal sources, and venom from Hymenoptera species, and related allergens sources are often used as homologous mixtures [ 8 9 ]. Manufacturing aspects in the production of extracts such as purity, quality, representativeness of the components as well as their storage are important to ensure effectiveness and safety in AIT [ 8 10 ]. Differences in the production of natural allergen extracts frequently occur and this heterogeneity in quality could generate different findings in the clinical and molecular outcomes of AIT [ 11 ].…”

“…The most frequently reported treatment-related adverse events (AEs) were mild-to-moderate oral pruritus and throat irritation as local reactions related to the sublingual administration. 14 In clinical trials, patients were solely treated with the tree SLITtablet. In real-life, since many patients are poly-allergic to tree and grass pollen, co-administration of tree-and grass-AIT is common.…”
