Safety of intravenous methamphetamine administration during treatment with bupropion

Abstract: Methamphetamine administration was well tolerated during bupropion treatment. There was no evidence of additive cardiovascular effects when the drugs were coadministered. This study provides initial evidence for the safety of prescribing bupropion for the treatment of methamphetamine abuse and dependence. The impact of bupropion treatment in patients who abuse larger doses of methamphetamine remains undetermined.

“…Even less is known about these effects with the intravenous formulation of methamphetamine administered in the human laboratory. The present study establishes the hemodynamic and cardiovascular effects and safety of experimenter-administered intravenous methamphetamine, thereby building upon other human laboratory data (Newton et al, 2005a(Newton et al, , 2005b. Notably, the administration of intravenous methamphetamine doses of up to 30 mg under controlled human laboratory conditions to methamphetamine addicts appears to be safe and only associated with prototypical increases in hemodynamic response.…”

“…Adverse events observed in this study were minor, with both active and placebo groups reporting similar magnitude and severity of events. In fact, bupropion treatment tended to blunt the cardiovascular effects of experimentally administered methamphetamine, with this effect reaching statistical significance for heart rate (Newton et al, 2005).…”

“…Before conducting this outpatient trial, we conducted a phase I safety study of the interactions between bupropion and methamphetamine (Newton et al, 2005). That study showed bupropion did not exacerbate the cardiovascular or behavioral effects induced by methamphetamine.…”
