Saccharomyces boulardii improves humoral immune response to DNA vaccines against leptospirosis

Abstract: The results suggested that Saccharomyces boulardii has an immunomodulator effect in DNA vaccines, mainly by stimulating the humoral response, which is often limited in this kind of vaccine. Therefore, the use of Saccharomyces boulardii as immunomodulator represents a new alternative strategy for more efficient DNA vaccination.

“…Nevertheless, this study demonstrated the feasibility of using a whole organism as a carrier of leptospiral proteins expressed at its surface to induce an immune response against leptospires after oral administration. The yeast Saccharomyces boulardi, administered in food pellets, and generally recognized as a safe (GRAS) organism, was recently shown to enhance the antibody and cytokine responses in Balb/C mice after 3 injections of DNA vaccines expressing LigA or LigB fragments [87]. However, no challenge was done with L. interrogans and it is not known whether S. boulardi improves protection against live bacteria.…”

“…Indirect ELISA using sera before and after immunizations was used to evaluate the IgG antibody levels as previously described [15]. Briefly, 96-well polystyrene microplates (CralPlast) were coated with 200 ng/well of rLigAni or rLigBrep diluted in carbonate-bicarbonate buffer 0.05 M (pH 9.6).…”

“…Another study investigated the immunomodulatory effects of Saccharomyces boulardii , a probiotic known for enhancing anti-inflammatory cytokine production and immune cell activation ( Silveira et al, 2017 ). In combination with DNA vaccines encoding leptospiral protein fragments, S. boulardii significantly increased antibody titres and upregulated IL-10 expression, particularly with pTARGET/ligBrep vaccination.…”
