S-Phase induced RNAs control ORC1 engagement to H2A.Z and firing of early DNA replication origins

Abstract

The mechanistic events leading to alterations of DNA replication in normal and diseased cells remain unknown. The present study uncovers the role of a new class of non-coding RNAs induced in early S-phase, able to activate and repress replication origins formation. The discovery of a RNA-mediated mechanism governing the replication of c-MYC locus offers novel strategies to control DNA replication by modulating transcriptional activity, and pave the way for future clinical applications to treat cancer and genetic disorders.
